Angiogenesis and mortality are associated with breast cancer, one of the most common tumors in women. Tumor angiogenesis is affected by exercise. This study examined the effects of 12 weeks of exercise on HIF-1α, mir-15a, BCL-2, and VEGF gene expression in BALB/c female mice. Forty BALB/c (two week’s age) female mice with a mean weight of 17±0. 21g were separated into control and treadmill aerobic training groups. Mice developed malignant after receiving 200μL of MC4-L2 cells subcutaneously. Running at 15-20m min-1 for 10 weeks was the aerobic activity. Afterward, mice were killed and tumor tissue RNA was extracted. HIF-1α, mir-15a, and VEGF gene expression in BALB/c female mice were measured using quantitative real-time PCR (RT-qPCR). After 12 weeks of exercise, miR-15a expression rose 2. 6 fold, whereas HIF-1α, Bcl-2, and VEGF gene expression dropped 3. 1, 2. 6, and 2. 4 fold, respectively (p<0. 05). Exercise can activate pathways that slow breast cancer progression. More research is needed to confirm these findings and other molecular pathways.

Introduction: Herbal medicines are commonly used by many people with diabetes in addition to standard treatment. Plants contain numerous known and unknown compounds that may exacerbate or ameliorate diabetes complications. Therefore, it is crucial to be aware of the side effects of these herbs before prescribing them. This study aimed to investigate the effects of hydroalcoholic extracts of Securigera securidaca (HESS) seeds alone and in combination with glibenclamide on the angiogenic/anti-angiogenic balance in streptozotocin (STZ)-induced diabetic rats. Methods: Groups involved in this animal study included diabetic and healthy controls, three doses of HESS, glibenclamide, and combination therapy. Serum samples were collected and analyzed for a vascular endothelial growth factor (VEGF), fibroblast growth factor 21 (FGF21), fetal liver kinase 1 (FLK-1), soluble fms-like tyrosine kinase 1 (sFLT-1), and transforming growth factor-beta (TGF-β). Results: Induction of diabetes increased VEGF, FGF21, and TGF-β serum levels and decreased circulating FLK-1 and sFLT-1 factors. Herbal extract, except TGF-β, had little effect on the above blood levels even at the highest doses. Glibenclamide was more effective than the highest dose of HESS in improving the vascular complications of diabetes. Combination therapy with the highest dose of HESS partly enhanced the glibenclamide effects. Conclusion: Compared with glibenclamide as a standard chemical drug, HESS had no significant effects on the blood levels of the pro/anti-angiogenesis factor in diabetic rats. Glibenclamide attenuated the levels of the biomarkers and its effects were somewhat enhanced in combination with the highest dose of HESS.

Introduction: Breast cancer is the most common cancer in women worldwide, and the triple-negative subtype is the most invasive, with limited therapeutic options. Since miRNAs are involved in many cellular processes, they harbor great value for cancer treatment. Therefore, in this study, we have investigated the anti-proliferative and anti-invasive roles of miR342 in 4T1 triple-negative cells in vitro and also studied the effect of this miRNA on tumor progression and the expression of its target genes in vivo. Methods: 4T1 cells were transduced with conditioned media of miR342-transfected Hek-LentiX cells. MTT and clonogenic assays were used to assess the viability and colony-forming ability of 4T1 cells. Apoptosis and invasion rates were respectively evaluated by annexin/7-AAD and wound healing assays. At last, in vivo tumor progression was evaluated using H&E staining, real-time PCR, and immunohistochemistry. Results: The viability of transduced-4T1 cells reduced significantly 48 hours after cell seeding and colony forming ability of these cells reduced to 50% of the control group. Also, miR342 imposed apoptotic and anti-invasive influence on these cells in vitro. A 30-day follow-up of the breast tumor in the mice model certified significant growth suppression along with reduced mitotic index and tumor grade in the treatment group. Moreover, decreased expression of Bcl2l1, Mcl1, and ID4, as miR342 target genes, was observed, accompanied by reduced expression of VEGF and Bcl2/Bax ratio at the protein level. Conclusion: To conclude, our data support the idea that miR342 might be a potential therapeutic target for the treatment of triple-negative breast cancer (TNBC).

Background and Purpose: Exercise and obesity alter angiogenesis in adipose tissue, resulting in large changes in metabolic processes. The aim of this study was to compare the effect of eight weeks of resistance training and high-intensity intermittent aerobic training (HIIAT) on the expression of microRNAs associated with adipose tissue angiogenesis and antiangiogenesis in rats fed a high-fat diet. Materials and Methods: In the experimental study, 60 six-week-old male wistar rats (mean body weight = 192. 40 ±,11. 35) were obtained from Pasteur Institute of Iran. The animals were randomly divided into six equal groups after transfer to the laboratory environment and one week of exposure to the new environment: 1. Normal diet (CO), 2. Normal diet + Resistance training (NRT), 3. Normal diet + HIIAT (NHT), 4. High-fat diet (HF), 5. High-fat diet + Resistance training (HFRT), 6. High-fat diet + HIIAT (HFHT) (n = 10 per group). The training groups performed their training protocol for eight weeks and five sessions per week. Resistance training included climbing a 1-meter ladder with a 90-degree slope and 46 steps. Initially, the weight attached to the rats’,tails was 30% of their body weight and gradually reached about 180% of their body weight in the final week. The exercises were performed in five sets of four repetitions. High-intensity intermittent aerobic training was started on the treadmill at an initial speed of 15 meters per minute, and in each session, one to two meters per minute was added to the treadmill speed, depending on the ability of the rats. During the Pabani week, the exercise reached six stages of three minutes of running at a speed of 40 meters per minute with a slope of 15 degrees. To evaluate the effect of resistance training and high-intensity intermittent aerobic training and high-fat diet (58% of calories in the form of fat) on miR-221, miR-222, miR-329 and VEGF variables, subjects in each group were killed 48 hours after the last training session. Sampling of mesenteric adipose tissue was performed immediately after killing the animals. Data analysis was performed using independent t-test, one-way and two-way ANOVA at the significant level (P ≥,0. 05). Scheffe post hoc test was used to compare the pairwise. Results: The miR-221, miR-222 and VEGF in the Normal diet + Resistance training, Normal diet + HIIT and High-fat diet + Resistance training groups a significant increase was observed compared to the control group (P ≥,0. 01). However, miR-329 was significantly higher only in the high-fat diet group than in the normal diet + HIIAT group (P = 0. 022). Conclusion: The results showed that resistance training and HIIAT by increasing miR-221, miR-222, and decreasing miR329 led to the activation of pathways that increased VEGF protein and ultimately increased angiogenesis in rats with a high-fat and normal diets. Therefore, these exercises are an important factor in angiogenesis and mechanism in adipose tissue, which may be able to cause significant changes in adipose tissue during this process. However, in order to confirm this point and generalize it to humans, more research is needed, especially on human specimens. Also, since the effect of resistance training on miRNAs was greater than HIIAT, these exercises can be a good alternative to HIIAT.
