Abstract:

Background: Polyhydroxybutyrate is a biodegradable plastic that has widespread application in various industries and in the field of medical equipment production. In E. coli, the most cost of Polyhydroxybutyrate production (PHB) is related to its extraction and purification processes. E gene of Phage Фx174 with conformation of membrane tunnel is used for lysing the hosted bacteria. This study was conducted to investigate the function of E gene in conformation of membrane tunnel and release of produced PHB in E. coli.Materials and Methods: In this study, after amplification with polymerase chain reaction (PCR), PHB operon was cloned by pUC18 while its expression controlled by operon promoter. E gene was amplified with PCR and cloned by pMR103 plasmid while it’s expression controlled by T7 promoter. Lysis process and conformation of membrane tunnel were evaluated by electronic microscope. Analysis of produced PHB was performed by gas chromatography and Sudan staining. Results: With activation of E gene and lysis of bacteria, optical density of specimen decreased. In the analysis of bacterial debris which achieved from E. coli medium by electronic microscope, we observed membrane pores in lysed bacteria. The results of gas chromatography and Sudan staining showed that PHB granules have been released to medium by conformation of membrane tunnel. Conclusion: Lysis E gene can be used for decreasing the cost and facilitating the extraction of PHB in E. coli that can be applied for manufacture of disposable medical and operating room equipment.

Abstract:

Background: Breast cancer is the most common type of malignan-cy in women. Despite dramatic achievements in the development of synthetic pharmaceutics, herbal drugs still remain as one of the leading therapeutic strategies for cancer. In the current study, we aimed to investigate the toxic effects of dendrosomal nanocurcumin on 4T1 cells derived from BALB/c mice breast tumors.Materials and Methods: For this purpose, MTT assay was exploited to evaluate viability of 4T1 cell line and mouse skin normal fibro-blasts (MEF) cells under exposure to nanocurcumin. Cultured cells were treated with different concentrations of nanocurcumin ranging from 5 to 45mM. Furthermore, the effect of nanocurcumin on cells was compared with doxorubicin as a frequently-used chemotherapeutic for breast cancer. Results: The results of MTT assay indicated that treatment with nanocurcumin leads to a significant reduction in the viability of cancer cells in a dose- and time- dependent manner with no significant effect on normal cells. The decrease in viability of cancer cells was 35, 25 and 15mM for 24h, 48h and 72h post-treatment, respectively. The data also shown that nanocurcumin exerts toxic effects the same as doxorubicin on cancer cells. Given less toxicity of nanocurcumin on normal cells in comparison with doxorubicin, this finding is of paramount importance. Conclusion: Dendrosomal nanocurcumin is able to eradicate cancer cells in a dose- and time-dependent mode. This nanodrug has toxic effects on breast cancer cells with no toxicity on breast normal cells. On the other hand, this therapeutic agent is as potent as doxorubicin in killing cancer cells. While compared with doxorubicin, this compound shows less toxicity on normal cells.

Abstract:

Background: Chronic neck pain is one of the common problems among computer users. The aim of this research was evaluation the effect of therapeutic exercises programs on the rate of chronic neck pain and disability in police staff in Lorestan Province.Materials and Methods: In this semi experimental study, thirty patients with chronic neck pain with the mean age: 35.73±5.68 years, height: 176.00±5.43 cm and weight: 72.9±10.61 and with at least six months history of neck pain were recruited by personal information questionnaire among computer users of the Police Headquarter of the Lorestan Province. The subjects were divided into two groups with 15 subjects. First group received therapeutic exercises by pamphlet, while the second one was the control group that did not receive any intervention. Neck pain and disability were measured by Neck Pain and Disability Scale at baseline and after 8 weeks intervention period. For analyzing data, first the differences between quantities of pre and post tests were measured, then evaluated by in-dependent t- test (P<0.05). Pearson correlation was also used to con-sider the relation between variables. Results: The results showed that neck pain and disability in experimental group were significant decreased compared with the control group (p<0.05). But Pearson correlation showed that there was no significant relationship between neck pain, disability and the subjects work experience (p=0.84, r=-0.037).Conclusion: According to the results of this study, it can be concluded that treatment by pamphlet would be useful for treatment of chronic neck pain. Consequently, it seems that applying this method was more helpful for the rehabilitation of patients with chronic neck pain and so it is highly recommended.
