Abstract: 

Background: The purpose of this study was to investigate the effect of aerobic training and Psilocybin supplementation on the expression of poly Adenosine diphosphate ribose polymerase (PARP) and dopamine receptor 5 (D5R) gene in heart tissue of methamphetamine-addicted rats.Methods: The present research was conducted experimentally with a post-test design with a control group. 30 female Wistar rats were randomly divided into five groups including healthy control; methamphetamine; Methamphetamine + aerobic activity; Methamphetamine + Psilocybin; Methamphetamine + aerobic activity + Psilocybin. Intraperitoneal injection of methamphetamine and Psilocybin was 10 and 0.02 mg/kg, respectively. The interventions of Psilocybin consumption and aerobic exercises were implemented for 4 weeks during the withdrawal period. Real-Time PCR method was used to measure gene expression in heart tissue. The Independent T, Man Whitney, and Kruskal Wallis statistical tests were used for data analysis.Findings: Methamphetamine use caused a non-significant (20%) increase in the expression of the D5R gene compared to the control group, but could not cause a significant change in the expression of the PARP gene. The use of the Psilocybin supplement and its combination with aerobic exercise produced a decreasing but insignificant effect on PARP and D5R gene expression in the heart tissue of female rats addicted to methamphetamine.Conclusion: Short-term induction of methamphetamine may not cause significant changes in the expression of PARP and D5R genes in the heart. Psilocybin supplement and its combination with aerobic exercise in the withdrawal period, even though it caused insignificant reduction effects in the expression of these genes, but the exact expression of gene changes requires further investigations.

Abstract: 

Background: During the outbreak of infectious diseases such as COVID-19, pregnant women and their fetuses are among high-risk groups, and vaccination is one of the most effective ways to prevent some infectious diseases. However, there is little information about the attitude of pregnant women, their husbands, and health service providers regarding receiving COVID-19 immunization. Therefore, the purpose of the present study was to explain the reasons for not receiving the COVID-19 vaccine during pregnancy.Methods: The present research was conducted in the framework of a qualitative approach and with the method of inductive content analysis in 2022. 29 participants including pregnant women, their spouses and, health service providers were selected with purposeful sampling and a maximum diversity participant selection strategy. The data collection tool was a semi-structured interview. Data analysis was done manually by the Granheim and Lundman method.Findings: The data analysis led to the emergence of 367 codes, 10 subcategories, and 3 main categories ("Incorrect performance of the health system", " Attitudes, opinions, beliefs " and "fears and uncertainties").Conclusion: Knowing the obstacles to vaccine injection helps implement effective interventions for optimal immunization coverage. It will be possible to inform pregnant women with the cooperation and strong role of midwives and gynecologists based on new and scientific documents and emphasizing the safety, effectiveness, efficiency, and role of the vaccine in reducing the mortality of pregnant mothers and even neonates until the acceptance of vaccination in similar crises will increase among pregnant women as one of the most dangerous groups in the society.

Abstract: 

Background: The purpose of this study is to investigate the relationship between the expression level of the important transcription factor POU5F1 in spermatogonial stem cells with age and to present the expression patterns of this factor in the mouse spermatogenic tube. Methods: In the present study, spermatogonial stem cells were isolated after extracting the testicles of two-week-old and sixteen-week-old adult mice purchased from the Pasteur Institute of Iran. The cells were cultured in a StemPro-34 medium containing specific growth factors. Also, after fixing and preparing the tissue, they were incubated with primary and secondary antibodies, and an immunohistochemical examination was done with a confocal microscope. Then, real-time PCR analysis was used to quantitatively investigate the expression of the POU5F1 transcription factor. Findings: While in the immunohistochemical analysis, high expression of the POU5F1 transcription factor was observed in the infant's testis, the number of POU5F1 positive cells in the seminiferous tubules of the adult testis was higher than that of the infant. Also, real-time PCR analysis showed that the level of POU5F1 gene expression in newborn SSCs was significantly (P < 0.05) higher than 16-week-old SSCs. Conclusion: The results can reveal the foundation of new dimensions in the relationship between epigenetic factors and the differentiation power of stem cells, which should be considered in research on the differentiation of spermatogonial stem cells into sperm cells.
