Abstract: 

Aim: This study was aimed to examine the effects of Stachys lavandulifolia extract (SL) on ethanolinduced testicular damages. Material and methods: In the current study, 24 male Wistar rats were divided into 4 groups: control, ethanol (5 g kg-10) and ethanol (5 g kg-1) + SL (500 mg kg-1) group and SL (500 mg kg-1). After the treatment for 4 weeks, mean seminiferous tubule diameter (MSTD) and Johnsen’ s mean testicular biopsy score (MTBS) criteria were used for histological evaluation. In addition, sperm parameters including sperm count and motility were analyzed. Results: Our data indicate that ethanol administration caused a significant decrease in the sperm count and motility in the ethanol group but SL ameliorated this reduction in SL + ethanol group. Moreover, both MSTD and MTBS values were decreased in the ethanol-treated rats. However, administration of ethanol together with SL resulted in significant improvements in both of these values. Conclusion: In conclusion, our study revealed that the reproductive toxicity was caused by ethanol may be prevented by SL treatment.

Abstract: 

Aim: In this study, the effects of recombinant human growth hormone and Gemcitabine alone and in combination with each other were investigated on the lung fibroblast cell lines. Material and methods: Human growth hormone at 10-400 ng/ml and Gemcitabine at 1-100 μ g/ml concentrations were treated with the human lung fibroblast cells and the analysis were performed with MTT assay, propidium iodide staining and scratch assay. Results: Studies of the cell cycle and MTT assay revealed the effect of growth hormone on the progression of the cell cycle and exit of G1 phase and the cell proliferation, and the inhibitory effects of the cell cycle and growth process by Gemcitabin compared with the control sample. The results of the scratch test indicated that the growth hormone at its effective concentrations caused increased the cell migration compared to control, while the Gemcitabin reduced the migration of the cell to the control. Conclusions: Using growth hormone in combination to gemcitabin during chemotherapy treatment, it seems that we can reduce the rate of the neem normal cells death. Since these cells are more sensitive to growth hormone´ s effect than cancer cells.

Abstract: 

Aim: An endogenous repairman of the central nervous system is possible via neural stem cells. Some researchers have suggested the neural protective of some medicinal herbs such as; Urtica dioica. The purpose of this study is creating an oxidative condition in the neural stem cell cultures and then treated the cells with methanolic extract of nettle leaves in vitro. Material and Methods: In this experimental study, the hippocampal neural stem cells extraction was performed by enzymatic digestion in newborn rats. The verification of these cells as nerve cells was carried out by the morphological and immunocytochemistry examinations. Before treatment, the cells were exposed for 24 hours to oxidative stress condition and then nettle leaf extract was added to the cell plates at of 2. 5, 5, 10, 20 and 40 μ g/ml and cells were maintained for 24 hours, separately. The final evaluation of the cell proliferation was performed by MTT viability test. The groups included; an experimental group that was treated with extract and the control group. Results: Neural stem cells had neural morphology and expressed nestin marker. Proliferation percentage of the neural stem cells was higher in the treatment groups than the control group. In addition, MTT assay results showed that the percentage of live cells in the treated groups increased, as the proliferation of the neural stem cells significantly increased (P<0/05) at 20μ g/ml. Conclusion: Methanolic extract of nettle leaf have neuroprotective effects and could adjust oxidative stress condition in vitro. It was able to improve the dysfunction of the central nervous system, following the production of free radicals.

Abstract: 

Aim: In this study, the toxicity effect of silver nanoparticles on colon cancer (HT29) was measured and evaluate the expression of the matrix metalloproteinase 9 (MMP9) genes. Material and methods: In this study, the synthesized AgNPs were characterized by the transmission electron microscopy (TEM) analysis. The cytotoxicity effect of silver nanoparticles against the HT29 cell lines was investigated using MTT assay. Also, MMP9 gene expression was measured using real time PCR technique. Results: The TEM result showed that the fabricated AgNPs were mostly spherical in shape and having an average diameter of 22 nm. The results revealed that AgNPs significantly decreased the viability of cells in dose-and time dependent manner (P<0. 001). Also, there was a significant relationship between the differences in concentrations. MMP9 gene expression levels in HT29 cells were decreased to 0. 4897 ± 0. 00679 (P<0. 001) compared with the control group. Conclusion: According to the results, the green fabricated AgNPs can be considered as a promising strategy for the treatment of colon cancer.

Abstract: 

Aim: In this study, the effect of insulin was investigated in induction of drug resistance to doxorubicin in MCF-7 breast cancer cell line. Material and method: MCF-7 cells were pretreated with 10 nM insulin for 48 and 72 hours, respectively. Then, different doses of doxorubicin (1, 5 and 10 μ M) were added for an additional 24 hours and cell viability was determined by MTT assay. Results: Doxorubicin had antitumor effects by reducing the cell survival in a dose-dependent manner. However, the addition of 10 μ M doxorubicin in the insulin-treated cells for 72 hours induced significant drug resistance (p<0. 01). Conclusion: The results revealed that insulin could significantly cause the doxorubicin resistance in a time-dependent manner in MCF-7 cells.

Abstract: 

Aim: The aim of this research was to investigate the effects of different concentrations of Azadirachta indica )neem( aqueous extract on the expression of the encoding genes of the two phenylalanine ammonialysis and peroxidase enzymes in tomato (Matin and Early Urbana cultivars) in the interaction with root knot nematode caused by Meloidogyne javanica. Material and methods: The tomato seedlings were inoculated in the 4-6 leafed stages with different concentrations of the neem extract and then incubated with the pathogen. The plants sampling were at six time points (0, 12, 24, 96, 192 and 288 hours) after pathogen inoculation. The gene expression of the defense enzymes was investigated using the Real-time PCR techniques. Results: In both cultivars, the gene expression of POX and PAL enzymes in the combined treatment of the pathogen nematode and the aqueous neem extract had increasing trend with significant difference with the control. In Matin cultivar, the increase of the gene expression was higher and faster than of Early Urbana cultivar, so that the expression of PAL and POX genes in this cultivar was 18. 2 and 14. 5 times more than the control. In the early Urbana cultivar, the highest rate of PAL gene expression was observed at 19. 2 hours and was 7. 5 times, and in the case of POX gene at 96 hours and 3. 9 times when it was compared with control. In greenhouse experiments, the nematode pathogenicity indexes showed a significant decrease compared with the Early Urbana cultivar. Conclusion: According to the results, in order to protect the plant against the invasions of the patients, the expression of the defensive genes should be increased at the earliest stages after the inoculation of the patient. Early increase in expression of genes in the seedlings treated with neem extract seems to be a justifiable phenomenon in the plant resistance to the pathogenic agent.

Abstract: 

Aim: This study was performed to evaluate the cytotoxic effect of Ocimum basilicum and Impatiens walleriana aqueous extract on the gastric cancer of AGS cell line. Material and methods: FRAP method was used to measure the antioxidant activity of Ocimum basilicum and Ipatiems walleriana extracts. After preparation of O. basilicum and I. walleriana aqueous extract, we measured the anticancer effect on growth of AGS cancer cells at 24, 48 and 72 hours by MTT. Results: The highest antioxidant activity of O. basilicum and I. walleriana was measured by FRAP method for water solvent and water bath extraction method, which in O. basilicum was 0. 778 ± 0. 11 mM and in I. walleriana 1. 30 ± 0. 03 mM. Based on the obtained data of the MTT test, the highest toxicity of the extract occurs 72 hours after adding the extract to the cells. The lowest amount of IC50 of the aqueous extract of O. basilicum was measured at 0. 012 ± 0. 002 mg / ml and in I. walleriana extract was 2. 87 ± 0. 001 mg / ml., which had a significant difference ( p <0. 05). Conclusion: The results of the anticancer study of the aqueous extract of O. basilicum and I. walleriana indicate that these plants have a cytotoxicity effect in the higher concentrations and need a long duration for their effects.

Abstract: 

Aim: In the current study, leaf anatomical structure and tissue cellular variation was evaluated in three populations of M. anisodon in Iran. Material and Methods: Two individuals were selected from each population. Leaves from middle part of the stems were fixed in F. A. A fixative and thin hand– cut sections were prepared of them. Then leaf samples were double stained and studied using a light microscope. Data were analysed using SPSS and MVSP softwares. Results: In all studied populations, leaves trans sections were dorsi– ventral. But, qualitative and quantitative traits of leaf differed among the populations. The ANOVA test did not show significant variations among most of the quantitative traits. Populations clustered separately in the UPGMA tree and also PCA and PCO plots. The population’ s arrangement pattern did not fit with geographical distances between populations. Conclusion: Similarity in the ecological conditions of habitats lead to anatomical structural likeness, and this case dose not related with geographical distance. Therefore, wherever the ecological conditions are similar, the likeness of leaf anatomical structure is very possible.
