Abstract: 

Leishmaniasis is a tropical parasitic disease that has become a major health challenge in many countries of the world. Not only has not been found any effective vaccine or treatment for the disease eradication, but also the advent of drug resistance is also increasing. Therefore, it is vital to take a precise attention to the physiochemical cycles of the Leishmania parasite and to identify its biochemical pathways. One of the most important biochemical pathways of host and parasite is the arginase and nitric oxide cycles. By using L-arginine, arginase plays an important role in the metabolic pathways, particularly in ornithine production, polyamines biosynthesis and cellular activities, including proliferation and cell survival. Furthermore, L-arginine, can act as a substrate for inducible nitric oxide synthase (iNOS ), which leads to the synthesis of nitric oxide (NO ), thereby activating the cellular immune system and clearing intracellular parasites. High Arginase activity reduces the parasite load inside the host cell, and since lymphocytes need L-arginine for their activity, its deficiency impairs the response of host immune cells. Also, parasites arginase alone can determine the fate of Leishmania parasite within the host cell. The aim of this study was to provide a comprehensive overview of various studies on the arginase activity of both parasite and host and its direct impacts on the immune system and pathogenicity of the Leishmania parasite.

Abstract: 

Background & objectives: Allium cepa has anti-cancer, anti-oxidant and anti-inflammatory properties due to flavonoid compounds. Allium cepa can prevent the onset and progression of cancer by removing free radicals. In this study, the preventive effect of Allium cepa in preventing the proliferation of breast cancer cells in BALB/c mice was evaluated. Methods: In the present study, Female BALB/c mice at 6– 7 weeks of age were subjected to the abdominal mammary gland subcutaneous injection with 5×10 viable 4T1 cells. Eight mice were placed in 3 groups: healthy group, the patient without treatment and the experimental group. The experimental group received Allium cepa root juice, three weeks before induction of disease. Daily, each mouse in the experimental group received 0. 1 ml / 100 g BW / day fresh Allium cepa root juice. Seven weeks after induction, the mice were exposed to deep anesthesia. During the test, the mice were weighed every other day and after the appearance of the tumor, by the end of the seventh week, the volume of the tumor was measured using digital caliper and the tumor mass was removed and weighed. The spleen was removed from the body and cultured in 1640-RPMI medium containing 10%, FBS, and ELISA test was used to measure IFN-γ and IL-4 levels. Data analysis was performed using SPSS software version 18. One-way variance analysis was used to assess the difference between the groups and, if it was significant, Tukey's post hoc test was employed to determine the differences between the groups. Also, p-value less than 0. 05 were considered as the level of significance for examining the hypothesis test and deciding whether or not to reject the hypothesis. Results: The weight of mice in all three groups increased and tumor weight and tumor volume decreased significantly in the experimental group compared to the patient group (sham 6 ) (p<0. 001, p<0. 0001 respectively ). In the experimental group, compared with the patient group, the levels of IFN-γ (p<0. 001) and IL-4 levels decreased significantly (p<0. 001). Conclusion: Based on the results of this study, the use of Allium cepa can prevent and reduce the growth and proliferation of cancer cells and disease progression in breast cancer mouse model.

Abstract: 

Background & objectives: Immunosuppressive drugs that are used for decreasing risk of acute rejection and renal graft loss can lead to reactivation of latent viruses for example BKV and JCV in either renal allograft or recipients. These viruses can lead to renal graft loss. The purpose of this study was to evaluate the quality and quantity of the genome of these two viruses in the renal recipients’ plasmas for early detection. Methods: In this retrospective descriptive study, at first, DNA extraction test was performed on 102 plasma samples of renal allograft recipients. And then, BKV and JCV DNAs were detected and quantified by Real-Time PCR. Results: Fifty four (52. 94% )BKV DNA positive and 26 (25. 50% ) JCV DNA positive were found in 102 recipient plasma samples. Linear range of measurements for BKV and JCV DNAs were within the range of 10 7-0. 596 copies/ µ l and 10 7-0. 528 copies/ µ l respectively. After calculation of genes amounts based on copy/ml in the plasmas, numbers and percent of positive cases were highlighted in the four categories. BKV and JCV DNA (Co-infection) were detected in 22 (21. 56% ) plasma samples. Conclusion: Real-time PCR is a quantitative and qualitative PCR method that can detect genome of any type of organisms and their amounts (even in trace amounts), so using of this method is very important for early detection of viruses which can cause diseases and graft rejection in renal transplant recipients.

Abstract: 

Background & objectives: In recent years, regarding the side effects of chemical drugs, the use of medicinal plants has increased due to their low side effects, low costs and effective compounds. The aim of this study was to investigate the anti-inflammatory, anti-nociceptive, and anti-pyretic effects of hydroalcoholic extract of Rosa canina L. fruit in male mice. Methods: In this experimental study, 120 male BALB/c mice weighing 23-30 g were used. In each test, the mice were divided into 5 groups (in each group, n=6), including control group, positive control and three experimental groups treated intraperitoneally with hydroalcoholic extract of Rosa canina L. fruit at doses of 100, 200 and 400 mg/kg respectively. The antiinflammatory and antipyretic activities were measured using xylene-induced ear edema and brewer’ s yeast-induced pyrexia tests, respectively. In addition, the antinociceptive activity was measured using the abdominal constrictions induced by acetic acid and formalin tests. The data were analyzed by SPSS statistical software and One Way ANOVA test. The level of significance was set at p<0. 05. Results: 200 (p<0. 05) and 400 mg/kg (p<0. 001) doses of extract reduced significantly inflammation. Doses of 100 (p<0. 05), 200 (p<0. 01) and 400 (p<0. 001 significantly reduced pain in the abdominal constriction test and dose of 400 mg/kg (p<0. 05) decreased the chronic pain in formalin test. Extract treatment did not reduce fever in any of the existing doses. Conclusions: The findings of this study suggest that Rosa canina L. fruit has antiinflammatory and visceral analgesic activity, which may be due to its antioxidant potential.

Abstract: 

Background & objectives: Antibiotic resistance in Vibrio cholerae is a crucial matter in the world. Objective of this study was the improvement of cholera surveillance by assessing the antimicrobial resistance pattern and bacterial resistance genes in V. cholerae O1 isolates, reffered to Iranian Reference Health Laboratory, in cholera outbreaks during 2012-2015. Methods: This study is a cross sectional-descriptive research. Antimicrobial susceptibility test (AST ) to 8 antibiotics was performed on 113 V. cholerae O1 isolates using E-test method. For all isolates, conventional PCR method was used to detect the presence of tetracycline resistance genes (tetA, tetB and tetC ) and the sulfamethoxazole-trimethoprim resistance genes (sul2 and dfrA1 ). Results: All isolates were sensitive to ampicillin, temocillin, ciprofloxacin and cefixime and 64% of strains showed intermediate susceptibility to erythromycin. The resistance rate of nalidixic acid, sulfamethoxazole-trimethoprim and tetracycline were 90%, 71% and 50% respectively. However, the frequency of multidrug resistant (MDR ) strains varied across the years. The frequency of resistance genes (tetA, tetB, tetC, sul2 and dfrA1 )were 70%, 34%, 58%, 66% and 70% respectively. Conclusion: AST should be used to determine the resistance profile at the beginning of a cholera outbreak and to monitor the resistance profile of circulating strains as part of surveillance of the disease. A prominent association was observed between phenotypic resistance to sulfamethoxazole-trimethoprim and presence of dfrA1gene. Determining the presence of resistance genes is necessary for understanding the epidemiology and routes of transmission of antibiotic resistance genes.

Abstract: 

Background & objectives: Type 2 diabetes mellitus is characterized by metabolic disorders and elevated blood glucose. It is one of the most common diseases in developed countries. Thyme is one of the herbs which have been used in some drug content due to its high antioxidant properties. Thyme can increase the activity of mitochondrial oxidative enzymes and affect metabolic and hematological indices due to its phenolic compounds. So, the aim of this study was to determine the effect of Thymus migricus extract supplementation on the metabolic, hematological indices and oxidative stress in type 2 diabetic rats. Methods: For this purpose, 24 Wistar rats (220-240g ) were divided randomly into three groups; group1: healthy control group (NC, n=8), group2: diabetic control group (D, n=8) that took fat diet for 2 weeks then were injected with streptozotocin (37 mg/kg ), and group3: diabetic+Thyme group (Th+D, n=8). Thyme hydro-alcoholic extract dissolved in distilled water to the desired concentration (400 mg/kg ) according to their daily water consumption (30 ml ). Then glycemic, blood and lipid indices were measured in the peripheral blood of the rats. Results: The results revealed that thyme supplementation significantly decreased the fasting blood glucose level (356. 35± 40 mg/dl to 261. 61± 35 mg/dl ) and HOMA-IR index (3. 52± 0. 30 mg/dl to 2. 2± 0. 33 mg/dl ). Also significantly decreased the lipid profile level of the Th+D group compared to those of the D group (p<0. 05), but the hematological parameters of the Th+D group compared to those of D group were not significantly changed. FRAP in Th+D group was not significantly different in comparison with the other two groups, but MDA was significantly increased in diabetic groups (p<0. 05). Conclusion: Based on the results, thyme supplementation by decreasing insulin resistance and fasting blood glucose can improve lipid parameters, but does not affect blood parameters and antioxidant indices.

Abstract: 

Background & objectives: Suicidal thoughts have been found to be associated with a wide variety of factors. A key factor is a history of childhood and family adversity. Previous studies have shown that troubled romantic relationships are associated with higher risk factors for mental health. Also, evidence suggests that those who have a secure attachment styles in childhood have fewer symptoms of psychopathology and higher psychosocial functioning throughout the lifespan (especially romantic relationship) compared to individuals with an insecure attachment style. The purpose of this study was to determine the role of emotional failure and attachment styles in predicting suicidal thoughts among university students of Azad university of Ardabil. Methods: In this study, descriptive and correlational design was defined as a research method. The statistical population of this study was all university students of the Ardabil Azad University, who studied in the academic year of 2018 (N=4231). The sample consisted of 80 people who were selected by purposive sampling. Beck's suicidal thoughts questionnaire, Simpson's attachment style questionnaire and Ross's love hit questionnaire were used to collect data. Data were analyzed using Pearson correlation coefficient and multivariate regression analysis. Results: The results of the research showed that emotional failure (r=0. 35) and unsafe attachment style (r=0. 23) had a positive correlation with suicidal thoughts, and secure attachment style (-0. 40) had a significant negative correlation with suicidal thoughts (p<0. 01). The results of regression analysis showed that 12 percent of the variance of suicidal thoughts can be predicted by emotional failure and 25 percent by attachment styles. Secure attachment style (β =3. 35), emotional failure (β =3. 35) and insecure attachment style (β =3. 3) were the most effective factors in the prediction of suicidal thoughts, respectively. Conclusion: Overall, the results of this study emphasized that attachment styles (secure and unsafe ) and severity of emotional failure are good predictors of suicidal thoughts among university students.

Abstract: 

Background & objectives: Nonsteroidal anti-inflammatory drugs (NSAIDs)-induced peptic ulcer disease is a common gastrointestinal disorder affecting many people worldwide. Rosa canina L. is a member of Rosa species that has been widely used as an herbal medicine in many countries. Rosa canina has therapeutic effects against various disorders through its antioxidant, anti-inflammatory and cytoprotective properties. The aim of the present study was to evaluate the potential beneficial effect of Rosa canina on indomethacin-induced ulceration in mice. Methods: Experiments were performed on male Swiss albino mice. Indomethacin (80 mg/kg, orally ) was used to induce gastric ulceration. The extract of Rosa canina fruits (200 and 400 mg/kg/day ) was given orally five days before the administration of indomethacin. Normal mice were used as a normal control group. Results: Macroscopic evaluation of gastric mucosa revealed that indomethacin administration (80 mg/kg ) produced apparent gastric mucosal ulcers, while pretreatment with Rosa canina (200 and 400 mg/kg ) effectively protected gastric mucosa against indomethacin induced ulcers. Rosa canina significantly increased the level of glutathione (GSH ) and decreased the levels of malondialdehyde (MDA ) and myeloperoxidase (MPO ) in the gastric tissues compared with those in the control group. Conclusion: Oral treatment with the Rosa canina fruit extract decreased the gastric ulcer induced by indomethacin through antioxidant and anti-inflammatory mechanisms.

Abstract: 

Background & objectives: Breast cancer is one of the most important causes of cancer deaths especially in developing countries. However, some synthetic compounds such as thiosemicarbazone and its metal derivatives can play a very significant role in reducing the death rate from cancer. The aim of this study was to investigate alterations in the expression of BRCA1, p53 and Bcl-2 genes in breast cancer cell line. Methods: In this study, the effect of pyrazole derivative of thiosemicarbazone (T ) as well as T-bound iron oxide magnetite nanoparticles (T/ Fe3o4 np) were investigated on expression changes of Bcl-2, BRCA1 and p53 in MCF-7 cells. For this purpose, the toxicity of the compounds was first measured using the MTT reaction for 24 hr. Then, the mRNA expression levels were evaluated using real-time PCR reaction. Results: The results showed a high toxicity for the nano-pyrazole derivative of thiosemicarbazone magnetite so that; the fatality rate for treated thiosemicarbazone magnetite derivative cells was much higher than the compound without any nanoparticle. Also, real-time polymerase chain reaction analysis of gene expression indicated a high effect of the derivative on increasing the expression levels of BRCA1 and p53 genes and also decreasing the Bcl-2 gene expression of Bcl-2. Conclusion: The use of anticancer compounds like thiosemicarbazone which bound to iron oxide nanoparticles would increase the toxicity of the anticancer compounds along with enhancment of delivery of them with high solubility to target tissue.

Abstract: 

Background & objectives: Local systemic filling pressure measurement can be used to determine other hemodynamic parameters such as venous return and the stressed volume. The aim of the present study was to evaluate the relationship between local systemic filling pressures and central venous pressure. Methods: In this study, candidate patients of cardiac surgery in Imam Khomeini hospital of Ardabil, in the first half of the 1397, were chosen and studied by their satisfaction, then an intra-arterial catheter inserted to the upper limb, a tourniquet closed to the same arm. The arterial pressure measured during the surgery. Central vein pressure was also measured simultaneously. Finally, other variables obtained from transesophageal echocardiography and demographic data were recorded in each patient's checklist. Results: 43 patients (71. 7% ) were man and 17 (28. 3% ) were women. The Mean CVP, MSFP, CO, and CI were 8. 3± 4. 5 mmHg, 17. 0± 6. 1, 3. 27± 1. 1, and 1. 89± 0. 7, respectively. Ten patients (16. 7% ) had mild heart failure, 42 patients (70% ) had a moderate heart failure and 8 patients (13. 3% ) had severe heart failure. There were positive and significant relationship between CVP and MSFP (r=0. 478: p=0. 001). The EF status has no significant effects on CVP with SBP, DBP, MAP, CO and CI. There was significant and reverse correlation between PSF value with CO, and CI in severe heart failure (p<0. 05). Conclusion: we concluded that the local MSFP is a good and valuable substitute for CVP measurement, especially in severe heart failure.

Abstract: 

Introduction: Crimean-Congo hemorrhagic fever (CCHF )is a viral disease that is transmitted to humans through the tick bite, contact with blood and tissues of infected animals in slaughterhouses as well as infected patients in hospital. The mortality rate of CCHF is 20-50%. Case Report: The patient was a 20-year-old man, an Iranian farmer, who lived in Ardabil County. The epidemiologic survey showed that his right leg has been bitten by a tick in the past week. The onset of the disease was a sudden high fever, chills and muscle pain. Molecular analysis of blood by RT-PCR showed that it was positive for CCHF disease. Conclusion: Clinical and epidemiological findings are the most important strategies for diagnosis of CCHF disease.
