Abstract: 

Background & objectives: Diazinon (DZN) as an organophosphate pesticide widely used in agriculture is associated with reducing the antioxidant capacity of the cell. Use of thiol compounds such as N-acetyl cysteine (NAC) as an antioxidant decreases oxidative stress in the cells. The aim of this study was to investigate the effect of NAC as an antioxidant against DZN- induced oxidative stress in rat brain and heart.Methods: In present experimental study, male Wistar rats were randomly divided into four groups including: control group (corn oil as DZN solvent), DZN group (100 mg/kg), NAC group (160 mg/kg), and NAC+DZN, all of which were given intraperitoneally.24 hours after injection, animals were anesthetized by ether, and the brain and heart tissues were quickly removed. After tissues hemogenation, superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST) and lactate dehydrogenase (LDH) activities, as well as glutathione (GSH) and malondialdehyde (MDA) levels were determined by biochemical methods.Results: DZN increased SOD and GST activities and MDA level and decreased LDH activity and GSH content in brain and heart. Also, DZN increased CAT activity in the heart and increased it in the brain. Administration of NAC inhibited the change in these parameters.Conclusion: DZN through free radical production leads to enhanced membrane lipid peroxidation, depleted GSH content and oxidative stress induction in the brain and the heart.Administration of NAC as antioxidant decreases the DZN-induced oxidative stress by scavenging free radicals and GSH synthesis, but its protection is not complete.

Abstract: 

Background & objectives: Myocardial infarction (MI) isone of the most commonly heart diseases in all countries with the mortality rate of about 30%. Reperfusion of blocked coronary arteries plays an important role in reducing mortality and enhancing the quality of life of patients after acute MI.Methods: This clinical trial is to compare the efficacy of streptokinase administered intravenously in a two slow and accelerated groups: control group (slow intravenous infusion during 1 hour) and case group (half-hour accelerated intravenous infusion of streptokinase) with the sample size of 100 patients (divided randomly to 50 patients in each group). The data tool was a questionnaire, included the collected ECG changes before and after echocardiography and laboratory tests of patients. Data analyzed with SPSS software using chi-square test.Results: After reperfusion of the coronary artery, in the both case and control groups, elevation of ST segment has decreased statistically in two groups (60% and 84%, respectively). The complications rate following injection of SK in both case and control groups were 46% and 56%, respectively. The fraction in which 30% and 38% belong to cardiac complications, 32% and 22% to allergic symptoms and hematologic effects were 32% and 22%, in two groups, respectively and there was no statistically significant difference between two groups.Conclusion: According to the results of this study the accelerated (30 minutes) SK infusion can be fruitful for improving coronary reperfusion in patients with MI.

Abstract: 

Background & objectives: Recurrent spontaneous abortion (RSA) is defined by two or more consecutive miscarriages before 20 weeks of gestation. Adenosine deaminase (ADA) is an enzyme of purine salvage pathway and has two important isoenzymes ADA1 and ADA2. The adenosine deaminaseG22A polymorphism (ADA*2) increases the level of adenosine.Adenosine may play a protective role against recurrent spontaneous abortions, since it regulates blood flow into the uterus and placenta. In consideration of the effect of decreased enzymatic activity of adenosine deaminase G22A polymorphism on adenosine levels we evaluated the protective effect of ADA*2 allele against recurrent spontaneous abortions in north-west of Iran.Methods: A total of 100 women were recruited to form two groups. First one, with a history of recurrent spontaneous abortions (N=50), and the second one, without a history of abortions (N=50). Genomic DNA was extracted from peripheral blood with a commercial kit and PCRRFLP analysis was used to identify the G22A genetic polymorphism. Fisher's exact test and odds ratio values were used to compare the proportions of adenosine deaminase genotypes and alleles between women with and without a history of recurrent spontaneous abortion (p<0.05).Results: The frequency of homozygotes (AA) was 2% in control group, whereas no homozygote (AA) was found in the case group. The frequency of heterozygotes (AG) was 20% in control group and 8% in the case group (p<0.05). The frequency of homozygotes (GG) was 78% in control group and 92% in the case group (p<0.05). A significant increase in the frequency of AG genotype in controls (p=0.014, OR=0.348) relative to women with the history of RSA demonstrates the protective effect of AG genotype in controls.Conclusion: The data suggest that women carrying the G22A polymorphism (ADA*2 allele) and AG genotype which is associated with the lower enzymatic activity are better protected against recurrent spontaneous abortions.

Abstract: 

Background & objectives: Resistant microbial strains are a serious threat to public health in different societies. Among the extended-spectrum β-lactamases (ESBL) producing strains the Enterobacteriaceae family which is considered as the main factors producing urinary tract infections, have created many problems in treatment of this kind of infections. This study was conducted to determine the frequency of β-lactamase TEM-1 gene in Enterobacteriaceae isolated from urine samples in Ardabil city.Methods: Within 6 months, 400 urinary isolates of Enterobacteriaceae of inpatients and outpatients were collected in Ardabil hospitals and were identified by standard methods.Antimicrobial susceptibility of isolates was tested by disk diffusion method, and ESBL producer confirmatory test was conducted using combined disk. Finally, the frequency of β-lactamase TEM-1 gene in producing extended-spectrum β-lactamases strains was investigated using PCR.Results: From 400 isolates of Enterobacteriaceae, 150 cases (37.5%) were ESBL producing.PCR results showed presence of the TEM-1 gene in 69 cases (46%). The frequency of this gene in isolates ofEnterobacter (Aerogenes, Cloacae), Klebsiella (Pneumoniae, Oxytoca) and E. coliwas obtained to be 62.5%, 54.5% and 44.8%, respectively. Proteus mirabilis and Serratia marcescensstrains were lacking these genotypes.Conclusion: As regards the presence of TEM-1 gene, there is also increasing in other members of the Enterobacteriaceae family includingKlebsiella and Enterobacter in addition toE. coli, therefore sufficient identification of this strains is necessary to prescribe the right medicine.

Abstract: 

Background & objectives: Trichophyton rubrum isone of the most common pathogenic causes of dermatophytosis. One of the drugs prescribed for fungal infections is fluconazole which belongs to Azoles group of antifungal agents. Recently molecular typing methods have been developed for answering the epidemiological questions and disease recurrence problems. Current study has been conducted on 22 isolates ofTrichophyton rubrum obtained from patients randomly. Our aim was the investigation of correlation between genetic pattern and sensitivity to Fluconazole in clinical isolates ofTrichophyton rubrum.Methods: Firstly the genus and species of isolated fungi from patients have been confirmed by macroscopic and microscopic methods. Then, the resistance and sensitivity of isolates against drug have been determined using culture medium containing defined amount of drug. In next step fungal DNA has been extracted by RAPD-PCR (random amplified polymorphic DNA) with random sequences of 3 primers.Results: Each primer produced different amplified pattern, and differences have been observed in genetic pattern of resistant and sensitive samples using each 3 primers, but there was no bond with 100% specificity.Conclusion: The 12 sensitive isolates which didn’t grow in 50μg/ml concentration of drug, also had limited growth at the lower concentration of drug. Ten resistant isolates which grew in 50μg/ml of drug, also showed resistant to lower concentration of drug. There are differences in genetic pattern of resistant and sensitive samples. RAPD analysis for molecular typing of Trichophyton rubrumseems to be completely suitable.

Abstract: 

Background & objectives: Hair follicle stem cells (HFSCs) are multipotent and various types of HFSCs were introduced. HFSCs separation using cell surface markers is one of the interesting strategies in the replacement of old methods. In this study, we used magnetic activating cell sorting (MACS) to separate HFSCs.Methods: In this study, HFSCs were isolated from Balb/c mice and dissected under an invert microscopy, and bulge area isolated and the bulge cells cultivated about 14 days. The CD34 positive cells isolated using CD34 monoclonal antibody and magnetic activated cell sorting system (MACS), then the cells incubated in DMEM/F12 and 10% FBS. The CD34 positive cells counted using a neubauer slide and evaluated under a fluorescent microscopy.Results: Here, we isolated CD34 positive cells using MACS and 12±1.04% of HFSCs were CD34 positive and we found that, CD34 positive cells survived during 7 days cell culture in vitro.Conclusion: The results show that MACS is useful to increasing density gradient of cells in vitro.

Abstract: 

Background & objectives: In recent decades, nanotechnology has been developing in medical field, and most of the nanoparticles are highly valuable in hygienic care. Vulvovaginitis is an infectious disease giving rise to problem in genital tract andCandida albicans is the main cause of Vulvovaginitis.Methods: In this study, laboratory effect of gold nanoparticles has been investigated on Candida albicansisolates obtained from patients with Vulvovaginitis. Candida spp. isolates were obtained from 200 patients referring to medical diagnostic laboratory in Isfahan city during 2013, and identified using mycological methods including germ tube, clamydoconidia formation, and culturing on chrom agar media. Antifungal effect of gold nanoparticles was evaluated in concentration of 100, 50, 25, and 12.5 ppm on Candida isolates using well diffusion and microdilution methods. Fluconazole was used as a positive control and results were analyzed by SPSS15 software using Mann-Whitney, Kruskal-Wallis and average differences analysis tests.Results: The most averages of clear zone diameter for gold nanoparticle were 18, 15, 12 millimeter, respectively. Minimum inhibitory and fungicidal concentration of gold nanoparticle was determined 6.25, 12.5 ppm and for fluconazole were 50.25±19.48 and 100.50±38.96, respectively. Based on obtained results antifungal activity of gold nanoparticle was dependent to concentration.Conclusion: In current study, inhibitory effect of gold nanoparticles was evaluated against microorganism. Although the laboratories findings are promising, more investigation should be conducted for therapeutic standardization.

Abstract: 

Background & objectives: Yersiniosis is created by Yersinia enterocolitica O: 8 and causes problems in the world especialy in cold and mild countries. The purpose of this study is to evaluate the immunogenicity ofYersinia enterocolitica O: 8 oligopolysaccaride (OPS) conjugate to diphtheria toxoid (DT) as a vaccine candidate.Methods: After cultivation of bacteria, the LPS were isolated by modified hot phenol method.Then dialysis and concentration were done and the OPS were extracted by acetic acid 2%. To conjugate with diphtheria toxoid, ADH was used as a spacer molecule and EDAC as a linker.Conjugate was purified by gel filtration. Then 4 groups of female BALB/c mice were selected (15 mice in each group). Injection was performed intraperitoneally in three doses with two weeks interval. Then serum samples were collected and antibody response against OPS was measured by indirect ELISA method for detection of total IgG, IgA, IgM, IgG1, IgG2a, IgG2b and IgG3.Results: After second and third doses, OPS-DT recieved group showed significant increase in all types of antibodies titer in anti-OPS in comparison to group that recived nonconjugated OPS. The increase in titer of antibodies was as: OPS-DT>OPS>DT. A remarkable increase was shown in total IgG and IgM titers (with total amount of 3204 and 670, respectively). In IgG1 subclass the amount was 920 and in other subclasses of IgG (IgG3, IgG2a and IgG2b) the amounts were 910, 110, and 99, respectively.Conclusion: The results shows that OPS of Yersinia enterocolitica O: 8 increases the anti-OPS antibodies in the form of conjugate with diphtheria toxoid and could be considered as an appropriate vaccine candidate.

Abstract: 

Background & objectives: Substance abuse is one of the worst humanitarian issues in recent years which undermine the base and foundations of human society. Its prevention requires the application of multiple theories in various disciplines together with diverse methods and techniques. Several studies have been emphasized on the role of personal and familial variables as risk factors for substance use. However, this study was done in order to predict drug addiction attitude in adolescents according to the family training risk factors to prevent substance abuse and to design a model for the prevention of addiction.Methods: This study is a descriptive and survey research performed on a sample of 373 male and female students selected randomly among the five high school students in Ghaemshahr city. Then a questionnaire including parenting styles, attitude to addiction and social problem solving skill as well as a socioeconomic questionnaire distributed among the students. For data analysis, the statistical method of descriptive statistics and path analysis has been used.Results: Results of this study have shown that the component of parenting styles has a direct and positive impact on attitudes to drug addiction. In addition, there was a direct and positive non-significant relationship between the adaptive social problem solving skills and attitude to drug addiction and also direct and negative significant relationship between the non-adaptive social problem solving skills on this attitudes. A direct and negative significant relationship was also seen between parenting styles and attitude to drug addiction.Conclusions: Based on the results of present study, the components of parenting styles have a direct and negative impact on attitudes to drug addiction. Also there is a direct and significant relationship between the components of non-adaptive social problem solving skills and the variable of social attitude in adolescents. But the component of adaptive social problem solving skill has a direct and non-significant impact.

Abstract: 

Background & objectives: Successful implementation of diabetes care can reduce acute and chronic complications of diabetes. This study was conducted to evaluate the quality of care according to a care scoring system among people with type 2 diabetes referring to the diabetes clinic in Ardabil.Methods: This cross sectional study was conducted in 2014 and a total of 300 people with type 2 diabetes in age range of 20-70 years old were included via convenience sampling. Data were collected by an interviewer using a structured questionnaire and checklist. Each patient was classified according to a quality of care scoring system into 3 categories: ≤10, 15-20, > 20. Statistical analysis performed by SPSS v.20 software and descriptive analysis used to display data as frequency tables and graphs. Analytical tests such as ANOVA, Kruskal-Wallis, Chi squared, and fisher exact test were also used to analyze the data.Results: The mean age of patients was 54.13 ± 9.13 and the majority of them were women (72.3%). The mean of BMI and HbA1c was 29.58 ± 4.77 and 8.93 ± 2.13, respectively. The mean of FBS was 191.18 ± 84.23 and the average length of disease was 7.74±5.89 years. The mean of quality of care score among participants was 23.20±9.99. About one-third of patients (29.7%) had a quality care score≤10, half of them (51%) had a score between 15-20, and only 19.35% had a score>20. There was an association between HbA1c, FBS, diastolic blood pressure, cholesterol and LDL with a quality of care score which was statistically significant (p<0.05).Conclusion: In this study there was a gap between the current quality of care of patients and recommended standards. Therefore in order to minimize the current barriers and problems it is recommended to implement an effective interventional program. In addition, using a quality care scoring system is a rapid, easy and valid method for evaluating diabetes quality of care.

Abstract: 

Background & objectives: Ovarian cancer is the most common female reproductive cancer which is caused due to the malignant transformation of ovarian cells. This type of cancer is the fifth most common cancer among women and the primary cause of cancer deaths in the world. Axin2 gene is a tumor suppressor gene of the Axin family in WNT cycle which is essential for embryonic development. WNT proteins in this pathway have important intermediary role in cell messaging and in primary and secondary development of the embryo.Axin2 gene is activated as a negative feedback to prevent excessive proliferation of cells with simultaneous activation of WNT messaging. The aim of this study was to find the frequency of mutation in rs1133683 region of exon 5 in Axin2 gene and its relation with the risk of ovarian cancer.Methods: In this case-control study, 100 patients with ovarian cancer together with equal number of same age as controls were collected from Imam Khomeini Hospital. DNAs were extracted from blood and tissue and then were investigated by PCR-RFLP. Data analysis was performed using software SPSS (version 19) using logistic regression.Results: The results of study of mutation in rs1133683 region of exon 5 in Axin2 gene between two groups of case and controls indicated that there is no significant association between CT genotype with ovarian cancer (OR=1.26, 95%CI; 0.70-2.27, p=0.43). Also no association was observed between TT genotype of Axin2 gene and ovarian cancer risk (OR=1.56, 95%CI; 0.49-4.96, p=0.44).Conclusion: Study of mutation in rs1133683 region showed that there was no association between TT genotype carriers of Axin2 gene and the risk of ovarian cancer.
