IDENTIFICATION OF SELF INCOMPATIBILITY ALLELES IN IRANIAN NATIVE AND SOME COMMERCIAL PLUMS USING ALLELE SPECIFIC AMPLIFICATION BY PCR

Abstract

 in this study, the S-RNase genotype of 34 plum genotypes from different area of Iran, five commercial cultivars and the Mirobolan cultivar (in total 40 genotypes) were determined by PCR amplification of the S-RNase gene using degenerate primers. A total of 23 bands, ranged 320-1830 bp, were amplified using EM PC2consFD/ EM PC3consRD primers that 14 alleles were already reported by other researchers and 9 bands were new. Twenty-three genotypes amplified two bands showing hetetrozigocity in this locus and the rest of them indicated only one band. According to the results Sc, 621 bp and Si alleles had the highest frequencies (30, 17.5 and 12.5 percent respectively) that Sc had highest frequency in Iranian plums (17.1%) in Velian of Karaj population. The S-RNase genotype of “Santa Rosa” was determined as SbSc that differed from that previously reported (ScSe). Also “Shiro” showed ShSi genotype that differed from that previously reported with only one alleles (Sf). The difference between observated and reported S alleles in “Santa Rosa” and “Shiro” cultivars may be because of mutation in this locus or the genotypes of these cultivars in this study is different from those reported. PaConsI-F/ EM PC5consRD primers produced 14 bands ranged 875-2495 bp in studied genotypes. Twenty-Five genotypes amplified two bands with these primers and the rest of them indicated one band. One genotype produced no band using both sets of primers. The results of this study showed that PCR can be a rapid and effective method for determination of S genotypes in plum GAMETOPHYTIC SELF-INCOMPATIBILITY, LOCAL PLUM GENOTYPES, S-RNASE ALLELES.
