Title

GENETIC RELATIONSHIPS BETWEEN ALMOND CULTIVARS AND GENOTYPES ASSESSED BY RAPD MOLECULAR MARKERS

Abstract

 In this research RAPD molecular markers were used in order to study genetic relationships of 39 native and foreign ALMOND cultivars and genotypes. Genomic DNA was extracted from young leaf tissues and PCR reactions were done using 20 selected primers 10 nucleotides with high polymorphism. Selected primers produced totally 297 bands, among them, 288 were polymorph and had more resolution that were scored and analyzed. Among the used primers, BD-04 and BD-10 showed maximum polymorphic bands (23 bands) and resolving power (13.64) respectively. The average resolving power of used primers was 7.05. Based on the similarity matrix, maximum similarity (0.78) was observed between ‘Yazad-444’ and ‘Yazad-60’ genotypes and the lowest similarity was observed between ‘Shale Qazvin’ and ‘Hybrid Holo-Badam’. Based on cluster analysis, cultivars and genotypes at 0.44 similarity distance were divided in to 8 groups that were according to their geographical origin and morphological characteristics. Cluster analysis efficiently and effectively showed the differences between the cultivars and genotypes. Based on the results, cultivars and genotypes including ‘Dir Ras-e-Savojbolagh’, ‘D-124’, ‘Nonpareil’, ‘Mirpanj-e-Tehran’, ‘Pakotahe-e-Taleghan2’, ‘Nu.11-10’ and ‘Barg dorosht-e-Hamedan’ in some of the important traits such as late flowering habit, nut and kernel characters, were better than the other cultivars and genotypes. The results proved that, there was a wide variation between studied ALMOND cultivars and genotypes that can be used in ALMOND breeding programs.
