INVESTIGATION OF DEGRADATION OF TOLUENE BY BACILLUS CONSORTIUM ISOLATED FROM TOLUENE CONTAMINATED SOIL

Abstract

 In this thesis the BIODEGRADATION of TOLUENE by a mixture of bacillus isolated from the contaminated soil was investigated. After sampling from the mentioned soil, the TOLUENE tolerant bacillus was identified, by culturing bacteria over blood agar, the bacteria which show b hemolyze surroundings was isolated. Afterward the surface tension tests have been employed for determination of BIOSURFACTANT producers. After the emulsification tests, the bacillus was detected by biochemical methods and molecular methods (DNA extraction, PCR test and sequencing test). Among 23 isolated bacillus stains, 3 stains (B12, B6, B3) have surface tensions lower than 40 mN/m which they are detected as the producers of BIOSURFACTANT. the emulsification ability of the 3 mentioned stains was 75%, 70%, 78% respectively. All of the three strains were B. subtilis. The sequence comparison 16srRNA of three strains shows the significant similarity among them, in that 3 sequences have about 6 different nucleotides.After reaching the optimum condition for BIOSURFACTANT production by the mixture of 3 strains (consortium), it was found that the best carbon source is TOLUENE and the best nitrogen source was yeast extract. In addition, the optimized salt concentration and pH was 2% and 6 respectively. At the mentioned conditions, the best production and consequently best BIOSURFACTANT production by BACILLUS CONSORTIUM have been achieved. The surface tension in the mixture of strains culture was considerably lower than those in the mono bacilli culture. In that in each culture of mono bacillus of B12, B6, B3 the surface tension was about 37, 33.5 and 35 mN/m respectively while the surface tension in the culture of consortium was 29 mN/m. The emulsification ability of the mixture of the 3 strains was about 85%. Using GC-MS, it was found that the TOLUENE removal by BACILLUS CONSORTIUM was about 99.91%.
