PRODUCTION, EXTRACTION AND ACTIVITY MEASUREMENT OF CLOSTRIDIUM BOTULINUM TYPE A NEUROTOXIN ON THE BASIS OF QUANTITATIVE PARAMETERS

Abstract

 The purpose of this study was to quantify the mouse behaviors, caused by botulinum toxin type A. Acid precipitation of the whole culture was prepared at pH=3.5 by addition of 3 M of H2S04 and then centrifuged at 10,000 rpm for 20 min, at 4°C. The pellet washed three times with distilled water and was homogenized in 100 ml of 0.1 M Sodium titrate buffer with pH=5.5 for one hour at 25°C. The sample was recentrifuged at 10,000 rpm for 20 min to obtain the supernatant which was considered to contain the toxin. Partial purified toxin was prepared by addition of 70% saturated ammonium sulphate to the sample solution. The precipitation was redissolved in 0.1 M Phosphate buffer with pH=6.5 and the solution were dialysed against the same buffer at 4°C. Protein concentration was examined by Bradfored method and then determined 1-2 µ.g/ ml. Parallel to concentration, the toxicity test was performed by intraperitoneal injection of 0.1, 0.2, 0.3 and 0.5 ml of toxin solution. In order to change the qualitative parameters to quantitative ones, movement responses of animal to different toxin concentrations was graded from one to five and the results were analysed. Therefore, with changing the qualitative parameters to quantitative ones, it could be possible to define the biological activity of Botulinum Toxin type A.
