IMMUNOCYTOCHEMICAL STUDY ON THE INDUCTIVE EFFECTS OF DEPRENYL ON DIFFERENTIATION OF BMSCS INTO DOPAMINERGIC NEURONS

Abstract

 Introduction: BMSCs can become a valuable cell source as an autograft for clinical application involving regeneration of the central nervous system. Recently, multipotential stem cells were identified in adult BONE MARROW STROMAL CELLS; these cells can generate glial and neuronal-like cells with appropriate treatment in culture and can differentiate into neuron-like and glial–like cells. The aim of the present study is to evaluate immunocytochemistry of BMSC which differentiated into neuron-like using DEPRENYL.Method & Material: Isolated BMSCs from adult rat were passaged for 5 times, and then using cytochemistry and immunofluorescent alkaline phosphatase reaction and fibronectin glycoprotein was assessed. The BMSCs in 5th passage treated with DEPRENYL after that compared with control group for assessment of BDNF (brain derived neurotrophic factor) expression and TYROSINE HYDROXYLASE reaction.Discussion & Conclusion: BMSCs differentiated into neuron– like cells when they are exposed to DEPRENYL at 10-8 M and expression TYROSINE HYDROXYLASE and BDNF. In conclusion, we have demonstrated that in during differentiation BMSCs into dopaminergic neuron which expressed BDNF using DEPRENYL as inducer.
