Abstract

The interaction of GUANIDINE HYDROCHLORIDE with thermophilic a-Amylase from B.subtilis and mesophilic a-Amylase from B.amyloliguefaciens has been investigated employing UV spectroscopy and kinetics measurments. Spectroscopic measurements were implemented at various temperatures (20-105oC) in 0.02M sodium phosphate at pH=6.9. Kinetics measurements were also used at different temperatures and pH (2-13). Thermal DENATURATION studies show higher stability for thermophilic in comparison with mesophilic type. The difference in Tm is 20oC. In the presence of GUANIDINE HYDROCHLORIDE, the stability of thermophilic enzyme increases, but the stability of mesophilic enzyme decreases. Thermodynamic parameters DHom and DSom increase for thermophilic enzyme but decrease for mesophilic enzyme. Specific activity measurements show more stability for thermophilic enzyme than mesophilic type.
