PHAGE TYPING OF THIRTY SYMBIONT RHIZOBIUM STRAINS ISOLATED FROM FIVE LEGUMES OF GILAN AND ISFAHAN PROVINCES SOILS

Abstract

 Thirty strains of symbiont RHIZOBIUM of five LEGUMEs were isolated from six different soils in Isfahan and Gilan provinces. Two phages of RHIZOBIUM leguminosarum biovar phaseoli (Phaseolus vulgaris L.) and RHIZOBIUM leguminosarum biovar viciae (Cicer arietinum L.) and two different phages of Sinorhizobium Meliloti (Trigonella foenum-graecum L.) were isolated and purified on their specific hosts for one year. They were used for phage typing of the thirty Rhizobia which were already isolated. Phages of RHIZOBIUM leguminosarum biovar phaseoli isolated from Jir-Gavabar area in Roudsar and Phages of RHIZOBIUM leguminosarum biovar viciae isolated from University of Isfahan region each had one CROSS REACTION. Phages of Sinorhizobium meliloti isolated from Najvan area of Isfahan and University of Isfahan region each had nine CROSS REACTIONs. Therefore, phages of RHIZOBIUM leguminosarum biovar phaseoli (Phaseolus vulgaris L) and RHIZOBIUM leguminosarum biovar viciae (Cicer arietinum L) phages can be used for identifying host strains among different strains of RHIZOBIUM isolated in the two provinces. Sinorhizobium meliloti phages can be used for differential diagnosis of RHIZOBIUM leguminosarum biovar phaseoli and RHIZOBIUM leguminosarum biovar viciae. Non-specific functions of Sinorhizobium meliloti phages indicated more extensive disruptive effects of them. Hence, by identifying relevant inhibitor agents, it may be possible to inhibit the invasivness of these phages biologically. Sensitivity or resistance of Rhizobia to BACTERIOPHAGEs was different, which could be correlated to the absence of specific receptors, lysogeny or presence of sym plasmid.
