ISOLATION AND PURIFICATION OF TOXIC FRACTIONS FROM THE VENOM OF SCORPION MESOBUTHUS EPEUS

Abstract

 Background & Aims: Scorpions in Khuzestan are mostly from Botideh family. VENOM of scorpions contains many biologically active compounds such as different TOXINs. These TOXINs are affected in Ionic channels in insects or mammals. In this study some TOXIN fractions from crude VENOM of Mesobuthus eupeus were isolated and purified.Materials & Methods: Six fractions were separated from crude VENOM by using gel filteration on sepha lex G-50.Discussion: From 816 mg protein of soluble crude VENOM which was applied on column CHROMATOGRAPHY, 815 mg protein was recovered in separated fractions. Of these six fractions, fraction III and IV were toxic to mice. The LD5o 0values of them were 0.4 and 0.46 mg/kg in mice respectively. Fraction III due to lower LDso or high toxicity was applied to a cation exchange column CHROMATOGRAPHY on CM-sephadex C-25 in the equilibration with 20 mM ammonium acetate buffer, pH 4.7. In this step, fraction III was separated into 20 distinct subtractions. Of these 20 subtractions, fractions III. 14 to III.19 were toxic to mice. Of 250 mg protein of fraction III that was applied on CM-sephadexC-25, 242.75 mg (97%) protein was recovered.Among the separated subtractions in this step, fraction III.14 had the highest toxicity with lower LD50 value. In final step for more TOXIN PURIFICATION the isolated fraction III.14 was applied to CM-sephadex C-25 column equilibrated and run with 20 mM ammonium acetate buffer, pH 6. In this step three distinct TOXINs were obtained in pure form. The LD50 values of purified TOXINs were determined, among those, TOXINs III.14.4 with a LD50 of 0.11 mg/kg in mice had the most toxicity effect. In this work three pure TOXINs were purified. The purified TOXINs can be used for providing antiserums against TOXINs and also to find more information about physiological and pathophysiological effects of these TOXINs.
