THREE-DIMENSIONAL CULTURE OF HUVEC CELLS

Abstract

ANGIOGENESIS is a highly complex physiological process which is an essential step in tumor proliferation, expansion and metastasis. It has been shown that anti-angiogenic agents help tumor therapy. It has also shown that Pectin and its modified forms can exhibit anti-mutagenic activity and suppress cancer metastasis and proliferation via inhabitation of ANGIOGENESIS with no evidence of toxicity or other effects. In the present study THREE-DIMENSIONAL CULTURE of HUVEC CELLS on collagen I to reach capillary tube formation is considered. We used the media of DU-145 CELLS (prostate cancer cells) which contains vascular endothelial growth factor (VEGF) to promote this process. DU-145 CELLS were cultured in Ham's F12/DMEM media with 1% FBS while HUVEC CELLS were maintain in a 20% FBS media. For ANGIOGENESIS formation HUVEC CELLS were grown on collagen I gel for 72h. Then the media of DU-145 CELLS were placed to their media and the tube formation was followed for a period of 48h. Our results show that however tube structure can form better in the absence of VEGF, the media of DU-145 CELLS support this formation greatly. In addition this formation can occur only on a THREE-DIMENSIONAL CULTURE and VEGF cannot stimulate tube formation in a common culture of HUVEC CELLS. We will investigate the effect of citrus pectin on ANGIOGENESIS by this method in our next research.
