EFFECT OF INTRA-VTA BUPROPION INJECTION ON MORPHINE WITHDRAWAL TEETH CHATTERING BEHAVIOR IN RAT

Abstract

BUPROPION as a Dopamine (DA) reuptake blocker is an antidepressant agent that is FDA approved for smoking cessation. DA neurotransmission in areas such as Ventral Tegmental Area (VTA) plays an important role in morphine dependence and withdrawal syndrome. Smoking addiction, like morphine dependence, is a synaptic plasticity with learning and memory basis. This study tested the short-term effect of intra VTA injection of BUPROPION on NALOXONE precipitated withdrawal syndrome teeth chattering sign. Wistar RATs were divided into 5 groups (control, non-dependent sham, dependent sham, 5 and 50 mg/μl/min BUPROPION-treated, n=6 in average). A guide cannula was implanted into VTA according to stereotaxic surgery under sterile condition. The sham groups received saline as drug vehicle but treated group received 5, and 50 mg/μl/1-2min of BUPROPION and control group had no injection. Morphine dependence was carried out under a standard protocol (i.e. intraperitoneal injection 3 times a day with incremental doses 15, 20, 25, and 30 mg/kg for 4 consecutive days). Withdrawal signs were precipitated with NALOXONE (1.2 mg/Kg, sc), observed continuously and registered on-line each minute. The behaviors were captured by 3 digital cameras from different views (30 min) for offline analysis. The teeth chattering was counted and analyzed within and between groups. The results showed that BUPROPION could dose-dependently reduce teeth chattering sign. BUPROPION could reduce teeth chattering 50% and 75% with doses 5 and 50 mg/μl/1-2min respectively. In conclusion, dopamine modulation by high-dose BUPROPION (50mg/μl/min) considerably reduces teeth chattering.
