Abstract

 Introduction and aim: In our previous studies using MICRODIALYSIS in the freely moving rats we showed the relation of stress with changes of noradrenaline (NA) and 5-Hydroxytryptamine(5-HT) release and synthesis in the rat brain. The present investigation aimed to study the role of brain 5-HT and NA systems on stress-induced behavioral responses.Material and Methods: Male Albino-Westar rats (200-300 g) were implanted with MICRODIALYSIS probes in the raphe, and lateral ventricle under chloral hydrate (500 mg/kg i. p.) anesthesia. Twelve hours after implantation animals were perfused at 2 ml/min with ringer solution. 5 min tali pinch is applied for control group. In the sham group ringer solution were perfused via MICRODIALYSIS probes while the TAIL PINCH (5 min) stress is performed. In one experiment, for the test group either Cd++ replaced with Ca++ in ringer solution or K+ (100 µM) added to the ringer solution. In another set of experiment, for the test group either clonidine (10 µM) or noradrenalin (20 µM) added to the ringer solution. The TAIL PINCH-induced behavioral responses were observed in all experimental groups. For validation of TAIL PINCH as a stressor, blood level of cortisone in control and TAIL PINCH groups were monitored. TAIL PINCH stress causes a variety of behavioral responses including gnawing, licking and aggression in rats. In the present study duration of these responses were observed at 5 min. Each group was compared. With the control animals. The statistical analysis was carried out using absolute data and either student paired t- test (within same group) or analysis of variance (ANOVA) statistical test (within the different groups).Results: The 5 minutes TAIL PINCH stress cause 231±11% increase in cortisol level. Local perfusion with potassium decrease the TAIL PINCH response to 37±6% of the sham group, while replacing calcium with cadmium in dorsal raphe nuclei increase TAIL PINCH response 110±2% of sham group. Local perfusion of conidian in lateral ventricle increase TAIL PINCH response to 112±3% of sham group.Conclusion: Data from the present study suggest an inhibitory role for 5-HT in those areas of the brain which may play a role in the stress-induced behaviors. Whereas in the other set of  experiment just we found a contrary role for brain NA in the stress-induced behaviors.
