Abstract:

Background: The erythrocyte binding antigen 175 kDa (EBA-175) gene is located on chromosome 7. It encodes protein that binds to specific receptor glycophorin A on the erythrocyte surface during invasion. It has a dimorphic nature (FCR3 and CAMP). This study was designed to determine the distribution of EBA-175 alleles of Plasmodium falciparum in the southeast of Iran. Methods: We used the nested PCR method with specific primers, which improves the two fragments of the EBA-175 gene. Sixty eight microscopically positive blood samples were collected from the infected falciparum malaria subjects in the southeast of Iran. Results: In this study which marks the first one in Iran, CAMP strains (714 bp) and FCR-3 strains (795 bp) were found in 14 (37.8%) and 23 (62.2%) in the originally Iranian subjects and in 10 (32.3%) and 19 (61.3%) Pakistani infected migrants respectively. Two migrant cases (6.4%) had mix CAMP/FCR-3 infection. Conclusion: The two fragments of dimorphic EBA-175 gene were observed and the FCR-3 allele was more prevalent in Iran. There was no significant correlation between one of the EBA-175 alleles and the subject group in the mentioned region. This distributional pattern should be considered in designing to control P. falciparum malaria in the region.

Abstract:

Comparative literature is one of the attractive and very valuable and useful trends in the field of literary criticism, and because the methods of research in comparative literature are different and numerous, there are different schools; In such a way that each researcher chooses the appropriate school or method based on his research method. With the emergence of the theory of reception in the last decades of the 19th century, more attention was paid to the role of the reader and the approach of central perception or reception was formalized. One of the research topics in the field of perception theory is the perception of Persian poetry by Arabic language writers and artists. In this context, Hafez Shirazi has attracted the attention of writers and researchers in the field of literature in other languages, and Arabic-speaking writers have a significant contribution in understanding Hafez's poetry. Among the writers who have paid special attention to Hafez's poetry and presented their views in the form of poetry and prose are Ebrahim Amin Al-Shwarabi and Ali Abbas Zulikha. In this research, two writers' interpretations of Hafez's Ghazal 175 are presented and analyzed and compared, and finally, the writer's opinion is presented as the final opinion. In general, it can be said that Al-Shwarabi's interpretation of verses is more comprehensive than Zuleikha's, but in terms of emotions and feelings, Zuleikha's interpretation is superior to Al-Shwarabi's.

Abstract:

Background: Breast cancer is the major cause of death from cancer among women around the world. Given the drug resistance in the treatment of this disease, it is very important to identify new therapies and anticancer drugs. Many studies demonstrated that hypericin could induce apoptosis in different cancer cell lines; however, the underlying mechanism is not well understood yet. Therefore, this study aimed to evaluate the anticancer effect of hypericin in two breast cancer cell lines, one with wild type P53 and the other with mutant P53. Methods: In this study, the MDA-MB-231 and MDA-MB-175-VII cell lines were treated with different concentrations of hypericin for 24 and 48 hours. The measurement of cell death was performed by MTT assay. The cell apoptosis rate was measured using annexin V/propidium iodide assay through flow cytometry. The level of expression in P21 and P53 genes was evaluated by real time PCR. Immunocytochemistry (ICC) analysis was performed for P21 (direct target for P53 protein) to confirm the results. Results: The results showed that hypericin could have dose-dependent cytotoxic effects on the MDA-MB-231 and MDA-MB-175-VII cell lines, and its cytotoxicity is much higher in the latter cells. According to flow cytometry results, 86% of MDA-MB-175-VII cells underwent apoptosis with IC50 dose of hypericin for MDA-MB-231 cells after 24 hours. Moreover, after 24 hours of exposure to hypericin with MDA-MB-231 IC50 concentration, the expression of P53 and P21 genes upregulated in MDA-MB-175-VII much more than MDA-MB-231 when both cell lines were treated with 24 hours IC50 dose of MDA-MB-231. The ICC analysis on P21 confirmed that by treating both cell lines with MDA-MB-231 IC50 dose of hypericin for 24 hours, this protein is overexpressed much more in MDA-MB-175-VII cells. Conclusion: The results of this study demonstrated that hypericin’ s apoptotic and cytotoxic effects on cancer cells may be mediated via P53 overexpression, cell cycle arrest and the subsequent apoptosis. Therefore, it is of great importance to consider that hypericin would have better impact on cells or tumors with wild type P53.
