Abstract: 

FT-SERS (Fourier Transform Surface Enhanced Raman Scattering) of ADRIAMYCIN and its complex with DNA is reported. It is shown that in agreement with previous Raman studies the interaction of ADRIAMYCIN with DNA takes place through an intercalation mechanism. The presence of a new band at 731 cm-1 suggests that ring D of ADRIAMYCIN is not involved in the intercalation process.

Abstract: 

In this study calf thymus chromatin was fractionated into active (S1and S2)and inactive (P2) chromatin. Then the interaction of an anthracycline antibiotic, ADRIAMYCIN, with these fractions was investigated employing absorption and difference UV/Vis spectroscopy and SDS and agarose gel electrophoreses. The results suggest that the binding of ADRIAMYCIN to the S1 fraction is slight but S2and P2 were considerably affected by drug action. In both cases the absorbances at 260 and 480 nm were decreased. As drug concentration is increased in the mixture, core histones are aggregated and coprecipitate with the chromatin. The extent of condensation in P2 is higher when compared to the S2fraction.

Abstract: 

Background: With respect to the known cardiac side effects of radiation and higher risk of these effects on patients with left breast cancer under radiation therapy following mastectomy, the present study was carried out in Radiotherapy-Oncology Department in Imam Hossein hospital in 2000 to determine the effects of radiotherapy on electrocardiography of the aforementioned patients. Materials and methods: 73 females referring for radiotherapy have enrolled in this clinical trial. They had undergone a modified radical mastectomy operation and received adequate chemotherapy protocol. Factors included in our study were menstruation state, ADRIAMYCIN chemotherapy protocol, and number of radiation fields. Results: The mean age of the candidates was 47 years. Short-term and long-term ECG changes in T wave and ST segment were reported 27% and 70%, respectively. In case of ADRIAMYCIN administration, ECG changes occur sooner with greater severity. With increasing the number of radiotherapy fields, the risk of ECG changes increased, however, the time of its development decreased. Postmenopausal females had experienced further ECG changes. Conclusion: Results have revealed short- and long-term ECG changes following the radiotherapy and chemotherapy. Thus, cardiac studies should be more seriously taken into account in the affected females.

Abstract: 

The aim of this study is to evaluate the effects of pravastatin on ADRIAMYCIN (ADR) -induced nephropathy and the mechanisms involved. Forty rats were divided into the following 4 groups: control, ADR (15 mg.kg-1, IP), ADR plus pravastatin (20 mg.kg-1 which was started 5 days prior to ADR injection), and ADR plus pravastatin (20 mg.kg-1 which was started 5 days after ADR injection). On day 20 after ADR injection, the animals were sacrificed. The results showed that administration of pravastatin decreased the levels of 24-h urinary protein (24-h UP), blood urea nitrogen (BUN), and creatinine (p<0.05) which had increased after the injection of ADR; in addition, pravastatin reversed structural changes seen in ADR group. Furthermore, pravastatin elevated mRNA and protein expression of nephrin (p<0.05) which had been reduced in ADR group. We conclude that pravastatin protects and treats renal injury induced by ADR.
