BACKGROUND AND AIM:THE THREE MAJOR TECHNIQUES FOR IDENTIFICATION OF BACTERIA ARE IOCHEMICAL TESTS, DNA SEQUENCING AND FATTY ACID PROFILING. THE MIDI SHERLOCK SYSTEM SING FAME ANALYSIS IDENTIFIES ALL OF THE AEROBIC BACTERIA IN ITS LIBRARY USING A STANDARD AMPLE PREPARATION TECHNIQUE, SO THERE IS NO NEED FOR UPFRONT BIOCHEMICAL TESTS OR A GRAM TAIN TO HELP DECIDE WHICH CARD OR TEST STRIP TO USE. BACTERIA ARE HARVESTED FROM A UADRANT OF THE STREAK THAT WILL MOST CLOSELY APPROXIMATE THE LOG STAGE GROWTH AND ROVIDE ADEQUATE CELLS FOR ANALYSIS. METHODS:SAPONIFICATION, METHYLATION AND EXTRACTION OR GC ANALYSIS WERE USED TO OBTAIN FULL BACTERIAL CELL FATTY ACID METHYL ESTERS (FAMES), OLLOWED BY A STANDARD MICROBIAL IDENTIFICATION SYSTEM ANALYSIS. RESULTS:SIMILARITY INDEX SI) IS A NUMERICAL VALUE WHICH EXPRESSES HOW CLOSELY THE FATTY ACID COMPOSITION OF AN NKNOWN COMPARES WITH THE MEAN FATTY ACID COMPOSITION OF THE STRAIN USED TO CREATE THE IBRARY ENTRY LISTED AS ITS MATCH. ACCORDINGLY, OUT OF FIVE TESTED ACINETOBACTER STRAINS, 2 ITEMS WERE CONFIRMED AS A.HAEMOLYTICUS AND 3 ONES AS A.GENOSPECIES. ALL OF THE ISOLATES WERE ALSO CHECKED BY PCR IDENTIFICATION TESTS AND THEY WERE SUBMITTED IN GENE BANK. CONCLUSION:ASIDE FROM THE METHODS SUCH AS PCR AND SEQUENCING TECHNOLOGIES, GAS CHROMATOGRAPHY (GC) FOR DETERMINATION OF CELLULAR FATTY ACID (CFA) COMPOSITION CAN BE USED AND THIS CAN SERVE AS AN ALTERNATIVE METHOD TO IDENTIFY THE ACINETOBACTER SPECIES AS WELL AS OTHER BACTERIA
