Abstract: 

Baculovirus expression system has been used successfully to over-express eukaryotic proteins in insect cells. This system uses a very strong viral promoter, AcNPV polyhedrin, for high level protein expression. Here a Baculovirus expression system was used to express the Mouse Gaq protein (mGaq) in SF9 insect cells. The recombinant protein was made at high levels and it was found in the cell membrane where it functions as a signaling molecule.

Abstract: 

Background and purpose: This study presents the effects of nitricoxide synthase inhibitor (L-NAME) on WIN55, 212-2 induced state-dependent memory of passive avoidance task, which were examined in mice.  Materials and methods: Mice were anesthetized with intra-peritoneal injection of ketamine hydrochloride, plus xylazine and then placed in a stereotaxic apparatus. Also, two stainless-steel annuals were placed 1 min above CA1. One-trial step-down paradigm was used for the assessment of memory retention in adult male NMRI mice. Results: Post-training intra-CA1 administration of WIN55, 212-2 (0.5 and 1 mg/Mouse), dose-dependent decreased the memory retrieval. The memory impairment induced by post-training administration of WIN55, 212-2 (1mg/Mouse), was restored by pre-test administration with the same dose of drug (1mg/Mouse, intra-CA1). Single intra-CA1 administration of L-NAME (0.3, 1 and 3 mg/Mouse), 5 minute pre-test could not alter memory retrieval. Also, in animals in which retrieval was impaired due to post-training administration of WIN55, 212-2 (1mg/Mouse), pre-test intra-CA1 administration of L-NAME (0.3, 1 and 3 mg/Mouse) 24 hours after training, could not restore memory retrieval. Furthermore, in animals which received both post-training (1mg/Mouse) and pre-test injection of WIN55, 212-2 (1mg/Mouse), the injection of L-NAME (3 mg/Mouse, intra-CA1), 2 minute before pre-test administration decreased retrieval. Conclusion: These findings may demonstrate the involvement of NO in state-dependent memory induced by intra-CA1 administration of WIN55, 212-2.

Abstract: 

The animal health monitoring is required to issue health certificates. The viral hepatitis virus is one of the most important infectious agents in mice breeding colonies. This research used RT-PCR to identify contaminations to Mouse hepatitis virus. 18 out of 29 specimens were found to be infected, a prevalence of 62%. PCR product was purified and sequenced. Phylogenetic analysis revealed that the identified strain in this study was closely related to a strain reported from France. In the conventional system, contamination with different infectious agents is inevitable, thus it is better to replace the contaminated colonies with clean animals...

Abstract: 

مقدمه: در مطالعه حاضر تاثیر تزریق داخل مغزی (i.c.v.) آگونیست و آنتاگونیست های رسپتورهای NMDA بر یادآوری با استفاده از روش ذیل مطالعه شده است. همچنین این داروها همراه مورفین تجویز شدهاند تا تاثیر تقویتی یا مهاری آنها بر اثر مورفین در یادآوری (یادآوری وابسته به وضعیت) بررسی شود. روشها: از موش سوری نر در محدوده وزنی 25 تا 35 گرم به عنوان حیوان مورد آزمایش و از دستگاه استرئوتاکس جهت تعیین دقیق محل کانول گذاری در مغز (داخل بطنی) استفاده گردید. یک هفته پس از جراحی و کانول گذاری با استفاده از روش (Two by-two factorial design) و متد step-down passive avoidance (اجتنابی غیرفعال)، یادآوری و مهار یادآوری با استفاده از داروهای ذکر شده مورد بررسی قرار گرفت. نتایج: 1- تزریق زیر جلدی مورفین 30 (5 mg/kg, s.c.) دقیقه قبل از آموزش (Pre-training) در مقایسه با گروه کنترل موجب مهار یادآوری و تزریق همان دوز از مورفین 30 دقیقه قبل از آزمون (Pre-test) موجب ایجاد یادآوری گردید. 2- تزریق داخل مغزی NMDA (10-3, 10-4, 10-5 mg/Mouse, i.c.v.) و یا L-glutamate (10-3, 10-4, 10-5 mg/Mouse, i.c.v.) قبل از آزمون در مقایسه با گروه کنترل تغییری در یادآوری (Retention latency) ایجاد نکرد. اما مهار یادآوری ناشی از تزریق زیر جلدی مورفین (5 mg/kg, s.c.) را برگشت داد. 3- تزریق داخل مغزی DL-AP 5 (1, 3.2, 10 mg/Mouse, i.c.v) و یا MK-801 (0.5, 1, 2 mg/Mouse, i.c.v) قبل از آزمون در مقایسه با گروه کنترل باعث مهار یادآوری گردید و Retention Latency را کاهش داد. نتایج نشان داد تزریق همان دوز از DL-AP5 و یا MK-801، مهار یادآوری ناشی از تزریق زیر جلدی مورفین (5 mg/kg, s.c.) را در مقایسه با گروه کنترل افزایش داد. 4- تجویز توام NMDA (10-3, 10-4, 10-5 mg/Mouse, i.c.v.) و یا L-glutamate (10-3, 10-4, 10-5 mg/Mouse, i.c.v.) و مورفین (5 mg/kg, s.c.) قبل از آزمون برگشت یادآوری ناشی از مورفین را افزایش میدهد. 5- تجویز توام DL-AP5 (1, 3.2, 10 mg/Mouse, i.c.v.) و یا MK-801 (0.5, 1, 2 mg/Mouse, i.c.v.) و مورفین (5 mg/kg, s.c.) برگشت یادآوری ناشی از مورفین را کاهش میدهد. نتیجه گیری: نتیجه گیری این که به نظر میرسد رسپتورهای NMDA در یادگیری اجتنابی غیرفعال وابسته به وضعیت مورفین در موش سوری میتواند دخالت داشته باشد.

Abstract: 

Human implantation and placentation need the direct contact of fetal with maternal tissues. The complex of maternal and fetal factors involve in maintain of pregnancy until delivery. The working on the human samples is problematic according to the ethical matters thus the Mouse models have contributed greatly to our understanding of the physiological and molecular events necessary for the process of implantation. Mouse models were suitable for analysis and understanding the physiological changes associated with implantation. Additionally, these models have ability to control uterine physiology through exogenous stimuli, and the ability to manipulate gene expression. Some investigations have focused on the understanding of the molecular mechanisms which regulate uterine receptivity and implantation. For this propose some ways introduce to induce failure In implantation or abortion such as heat stress, mechanical vibration, administration of some chemicals, and usage of proven aborted strains.
