BACKGROUND: IN RECENT YEARS SPREAD RESEARCH HAS BEEN INITIATED WITH NEW ADVANCED DRUG DELIVERY SYSTEMS WITH BETTER DRUG CONTROL, PROLONGED ACTION AND OPTIMIZED THE SECURITY AND EFFICACY OF THE TREATMENTS [1]. THE INDIVIDUAL FEATURES OF CHITOSAN (BIODEGRADABILITY AND NON-TOXICITY), BIOACTIVE GLASS (REPAIR AND RECONSTRUCTION OF BONE TISSUE) AND TIO2 NANOPARTICLES (FAVORABLE BIOLOGICAL EFFECTS) HAVE INSPIRED US THE FEASIBILITY OF COMBINING ALL TO MAKE AN EFFECTIVE MATERIAL AS A LOCAL-ANTIBIOTIC DELIVERY SYSTEM. TETRACYCLINE HAS BEEN CHOSEN FOR ITS BROAD ANTIBIOTIC SPECTRUM AND ANTI-INFLAMMATORY ACTION AS A DRUG MODEL [2].METHODS: FOUR SAMPLES INCLUDING S520 BG (S1), BG/CHITOSAN (MOLAR RATIO 1: 0.1, S2), BG/TIO2 (MOLAR RATIO 1: 1, S3) AND BG/CHITOSAN/TIO2 (MOLAR RATIO 1: 0.1: 1, S4) WERE PREPARED. ESCHERICHIA COLI WAS USED AS A MODEL BACTERIUM TO EVALUATE THE ANTIBACTERIAL ACTIVITY OF TETRACYCLINE LOADED COMPOSITE (S5), AND OTHER SAMPLES TO ASSAY INHIBITION ZONE OF BACTERIUM CULTURED [3]. FOR DELIVERY ASSAYS, ONE PELLET OF S5 WAS IMMERSED IN PHOSPHATE BUFFER SOLUTION (PBS) AND PBS REMOVED WAS ANALYZED TO MEASURE THE RELEASED TETRACYCLINE AMOUNT. THE BIOACTIVITY OF S4 AND S5 TO FORM HYDROXYAPATITE WAS ASSESSED AFTER SOAKING IN SIMULATED BODY FLUID (SBF).RESULTS: FIG.1 (A) SHOWED THE TYPICAL XRD OF THE S4 POWDER MATCHED WELL WITH THE STANDARD PATTERN OF ANATASE TIO2. THE ANTIBACTERIAL ACTIVITY OF SAMPLES IMPROVED IN COMPOSITES (S1, S2 AND S3) AS COMPARED TO S4 AS SHOWN IN FIG.1 (B). THE TYPICAL SEM IMAGE OF S4 AFTER IMMERSION IN SBF FOR 3 WEEKS IS SHOWN IN FIG.1 (C) ILLUSTRATING DEPOSITION COMPOSED OF HYDROXYAPATITE.
