Luciferase is the key enzyme that catalyzes the oxygenation of luciferin, generating energy-rich peroxidic intermediates, emitting a photon of visible light. Inositol 1, 4, 5- trisphosphate (IP3) plays a vital role in the control of a large number of cellular processes.Alteration in its content has been observed in metabolic disorders. Hence, development of high-throughput screening systems to monitor temporal changes of IP3 is essential for screening of new potential therapeutic compounds. Toward a sensitive, rapid and simple method to measuring IP3, we describe the development and application of a luciferasefragment COMPLEMENTATION strategy, which converts the ligand-induced conformational changes to light. In order to create a good performance sensor for IP3 monitoring, IP3 binding domain (IP3BD) of IP3R that consist of all the specific binding site of IP3 was selected as a ligand binding domain. The NLuc of firefly luciferase is fused to the Nterminus of IP3BD whiles the CLuc of luciferase is attached to the C-terminus of IP3BD (NLuc-IP3BD-CLuc). A flexible peptide linker (G4S) 2 was inserted between the fragmented luciferase and IP3BD to assist in proper protein folding and luciferase COMPLEMENTATION. According to the results, the screening time was very fast and maximum response was obtained up to 11-fold higher than background. Moreover, the designed biosensor was able to monitor release of IP3 upon induction by different inducer like Bradykinin and ATP. The current biosensor not only provides a specific IP3 detector in vitro, but also facilitates to monitoring of the response of IP3 in living organisms.

For a compact K, a necessary condition for C(K) to have the Controlled Separable COMPLEMENTATION Property is that K be monolithic. In this paper, we prove that when K contains no copy of [0,ww] and the set of points which admit a countable neighborhood base is a cofinite subset of K, then monolithicity of K is sufficient for C(K) to enjoy the Controlled Separable COMPLEMENTATION Property. We also show that, for this type of compacta K, the space C(K) is separably extensible.

We searched for novel Chinese hamster ovary (CHO) cell mutants defective in peroxisome biogenesis by an improved method using peroxisome targeting sequence (PTS) of Pex3p (amino acid residues 1-40)-fused enhanced green fluorescent protein (EGFP). From mutagenized TKaEG3 (1-40) cells, the wildtype CHO-K1 stably expressing rat Pex2p and of rat Pex3p(1-40)-EGFP, numerous cell colonies resistant to the 9-(10-pyrene) nonanol/ultraviolet treatment were grown. These colonies were examined for intracellular location of Pex3p (1-40)-EGFP. By this method, we have isolated one CHO cell mutant, ZPEG403, which was found to belong to COMPLEMENTATION group G (CG-G). Expression of the human peroxin, Pex3p cDNA encoding a 373-amino-acid peroxisomal membrane protein morphologically and biochemically restored peroxisome biogenesis, including peroxisomal membrane assembly, in ZPEG403 cells.Mutation and genomic DNA PCR analyses showed that, the dysfunction of Pex3p in ZPEG403, was due to one base (A) substitution in place of (G) in the first base of splicing site at the boundary of exon 6 and intron 6 of PEX3 gene, giving rise to remaining of all of intron 6, thereby inducing 81 bp insertion between positions 523–524 of PEX3 ORF, resulting in deletion of 200 amino acid residues from the C-terminus of Pex3p and a frame shift inducing both 18-amino-acid substitution and an early termination codon.

This question about the possibility of inventing a system more capable than humans in performing behaviors has always been on the table, even before the advent of powerful computers and artificial intelligence algorithms. Some linguists and philologists believed this process to be impossible. Triumph over the obstacles of linguistic barriers is one of the most important goals and tasks for developments in communication technologies. The idea of constructing an artificial communicative language, such as Esperanto, or the attempt at unifying the language for exchanging information has proven to be abortive due different reasons; therefore, the notion of automating translation process and using Machine Translation (MT) systems continues to be of consideration and reference. Machine Translation is one of the important achievements of artificial intelligence and computational linguistics. This field of knowledge contains the latest and greatest theories of applied linguistics and computer science. Today, we confide in the fact that the idea of exploiting artificial intelligence, and facilitating its application in multiple areas of science, has been a necessity; however, the implementations of this achievement were not provided by mechanics. This field was not a proper context for creating the reciprocal COMPLEMENTATION of linguistics, logic, statistics, and mathematics to revolve around it and could not provide the required agility to employ its achievements in accomplishing Machine Translation. On the other hand, with the advent of a phenomenon known as digital, not only did the procedures of formal logic turn into mathematical logic and that of the mathematics turned into modern mathematics, but also new trends were established in this regard and developed rapidly in linguistic researches properly relevant to digital contexts. After a brief introduction to the topic and its theoretical framework, this paper tries to determine how these achievements have been accomplished and intends to elaborate upon the future prospects with respect to the highlighted methods. While presenting his personal experiences in this area, the author of this paper emphasizes establishing a diverse institutional resource in modern linguistics. In his view, future achievements and accomplishments are due to developing the latest and fundamentally required data in linguistics as an interdisciplinary field of knowledge in order to gain practical achievements.

Let G= (V, E) be a graph. By directional labeling (or d-labeling) of an edge x=uv of G by an ordered n-tuple (a1, a2,…, an), we mean a labeling of the edge x such that we consider the label on uv as (a1, a2, …, an) in the direction from u to v, and the label on x as (an, an-1,…, a1) in the direction from v to u. In this paper, we study graphs, called (n, d) -sigraphs, in which every edge is d-labeled by an n-tuple (a1, a2, …, an), where ak Î {+,-}, for 1 £k£ n. In this paper, we give dierent notion of balance: symmetric balance in a (n, d) -sigraph and obtain some characterizations.
