DETERMINATION OF PHENOTYPIC CHARACTERISTICS AND HOST RANGE OF RHODOCOCCUS FASCIANS STRAINS THE CAUSAL AGENT OF FASCIATION ON ORNAMENTAL PLANTS IN SHIRAZ

Abstract

 In the present study 122 strains of RHODOCOCCUS FASCIANS were isolated from different plant and soil sources. Biological, morphological and nutritional features, susceptibility to antibiotics and host range of these strains were campared together with two isolates from North of Iran and a standard isolate (981011). Based on phenotypic features strains isolated from tobacco (Turkish & N. glutinosa) showed 98.6% similarity with isolate 981011 and nasturtium strains indicated lowest smilarity to standard isolate. Furthermore nasturtium isolates with similarity coefficient of 84.3 were the most similar to the North Iran's petunia isolate but geranium isolates with similarity coeficent of 78.5 had the lowest similarity to this isolate. All tested strains were resistant to amikacin, ciprofloxacin and chloramohenicol antibiotics. Nasturtium and N. glutinosa strains were more resistant to the antibiotics tested. The bacterium was 23% and 4% seed transmitted in diseased nasturtium and commercialdahlia, respectively.Host range of tested strains were different, and bean and cowpea infected with North Iran*s petunia and nasturtium strains showed stunting. Geranium strains were able to form LEAFY GALL in gladiolus and chrysanthemum plants. Chrysanthemum strains were unable to produce LEAFY GALL symptoms in plants tested, but reduced the size of bulbs in dahlia plants. None of the strains were able to produce disease symptoms in stock, carnation, rape, zucchini, tuberos and sesame. For isolation of bacterial strains from soil, D2 medium was more efficient than trap method. On the basis of morphological, physiological and biochemical features strains of R. fascians isolated from various hosts and soilsin Shiraz, apperently were homogeneous and more similar to the standard isolate, but were somewhat different from the North Iran's geranium strain.
