EVALUATION OF SEED VIGOUR OF FOUR ALFALFA CULTIVARS BY ACCELERATED AGING AND CONDUCTIVITY TESTS

Abstract

 Tow laboratory experiments were conducted to evaluate vigour of four ALFALFA cultivars of Yazdi, Charahyonge, Bami and Hamedani by ACCELERATED AGING and conductivity tests. In the first stage, seeds were exposed to 45°cand relative humidity of 100'percent for zero, 2,4, and 6 days. This seeds were then arranged in baches of 50 per petridish with 4 replications in a factorial design, and kept in germinator under temperature of 20±1 for 6 days. At the end of this period germination percentage, hypocotyle and radicle lengths were determined. Seed which had a radicle length of 2 mm were considered as germinated. For conductivity test, two replicats of 500 seeds were placed in 50 ml of distilled water and them expoed to 25° c in incubator for 24 hours. Electerical conductivity (EC) was then measured. Results showed that there was a significant difference between cultivars in terms of percentage of germination, radicle and hypocotyle length. Increased aging period reduced all this criteria. It was observed that 2 and 4 days were the best period for this type of test, because there was no difference between cultivars after 6 days. Amongst the cultivar Yazdi showed the highest and Hamedani the lowest vigour. In EC test a similar trend was observed as that of ACCELERATED AGING test. Therefore, EC test seems to be as effective in this type of experiment as ACCELERATED AGING test and if further clarification is made it is a rapid test for this purpose.
