Abstract

 The aim of this research was to micropropagate sour orange and lime by asexual method, using SHOOT-TIP CULTURE. Eight mm long shoot tips were separated and cultured on MS medium containing 0.1% agar and 3% sucrose. To stimulate shoot prolifiration, NAA was added at the concentration of 0.5 mg l-1 in all the media used. BA was used at concentrations of 0, 0.5, 1 and 2 mg l-1 and GA3 at concentrations of 0.5, 1 and 1.5 mg l-1. Phloroglucinol (PG) was also used at constant rate of 80 mg l-1. At the concentration of 0.5 mg l-1 BA, 87.5% of lime shoot-tips remained active and continued the growth. After sub culturing, highest shoot PROLIFERATION was also observed at this concentration of BA. GA3 and PG did not have any positive effect on shoot-tip PROLIFERATION of lime. Shoot-tips of sour orange had not any growth in all treatments. For root stimulation of lime, IBA and NAA were used at concentrations of 0.5, 1 and 1.5 mg l-1 in half-strength basal MS medium. The results showed that 1.5 mg l-1 IBA caused the highest ROOTING comparing to control and other treatments. In addition, 80% of plantlets were transferred and acclimatized successfully.
