A STUDY OF TEMPERATURE EFFECT ON CALLUS REGENERATION IN IMMATURE EMBRYO CULTURE OF BARLEY (HORDEUM VULGARE L.)

Abstract

 To examine the TEMPERATURE effects on CALLUS REGENERATION derived from IMMATURE EMBRYOs of BARLEY, three two-row genotypes and three six-row genotypes out of Iranian and Japanese BARLEY cultivars were selected from Cereal Collection Section of Agronomy and Plant Breeding Department, Agriculture Faculty of University of Tehran. Two weeks after anthesis, the IMMATURE EMBRYOs were cultured into M.S medium with 2 mg/lit 2,4-D. The CALLUSes were maintained in the darkness at 25 o C for approximatelyone month. Then they were transferred to REGENERATION medium, similar to the former medium but complemented with 1 mg/lit 2,4-D. Six TEMPERATURE treatments were employed in the experiment as follows: Control (25o C), treatment 2 (30 o C for 10 days), treatment 3 (5o C for 10 days), treatment 4 (5 days at 30o C followed by 5 days at 5o C), treatment 5(5 days at 5 o C followed by 5days at 30oC) and cycle treatment (every other day at 5o and 30o C respectively). The photoperiodism for all the treatments was 16 hours of light and 8 hours of darkness. TEMPERATURE effect treatments, with regard to shoot and root REGENERATION either, a significant difference or held the highest means in all the studied cases. Almost in all cases, heat treatment enhanced the number of regenerated shoots; CALLUS size and the length of the regenerated shoots, while cold treatment brought about an increase in the number of regenerated roots, a decrese in albino plants and an increase in the length of the regenerated roots. "ZarJo" cultivar showed the highest shoot and root REGENERATION as a result of the treatments 4 and 5, respectively.
