Introduction: Unilateral ureteral obstruction (UUO) is one of the causes of kidney injury. Oleuropein is known as one of the strongest natural antioxidants. The aim of this study was to determine the effect of complete left unilateral ureteral obstruction on the antioxidant system changes in the contralateral kidney in male rats with an emphasis on the protective role of oleuropein Materials and Methods: In this experimental study, 18 male Wistar rats weighing 200 to 250 g were used. The animals were divided into 3 groups of 6 which included control, UUO, and UUO + oleuropein. The left ureter was ligated for 3 days. In the treatment group, the animals received oleuropein (200 mg/kg) daily for 3-days from one hour after ureteral obstruction. At the end of the experiment, the right kidney was removed and weighed. Then, the malondialdehyde (MDA) concentration, glutathione peroxidase (GPx), and superoxide dismutase (SOD) enzymes activity levels were measured in the right kidney. Results: UUO significantly increased right kidney weight and MDA concentration, and decreased the SOD and GPx enzymes activity level in the right kidney of UUO animals compared to the control (p<0. 05). Oleuropein was able to reduce right kidney weight and MDA concentration in the UUO+oleuropein group compared to the UUO group (p<0. 05), and significantly increased the SOD and GPx enzymes activity level (p<0. 05). Conclusion: UUO induces oxidative stress in the contralateral kidney, and oleuropein administration can have a protective effect on the contralateral kidney by restoring the antioxidant system.

Introduction: Leber congenital amaurosis (LCA) is a vision loss disorder that begins in infancy. Differentgenes are associated with LCA. They usually have autosomal recessive inheritance. In the present study, the genetic basis of congenital blindness in two Iranian families was examined. Materials and Methods: Next generation sequencing technique was used for investigation of theunderlying mutations in patients with LCA. The Sanger sequencing was employed to validate the identified potential pathogenic mutations. Results: In the present study, two novel mutations in GUCY2D gene were detected in probands. Firstmutation was a frameshift mutation caused by single base deletion (c. 1264delC), and the second one was anonsense mutation (c. 2116C>T) resulting in stop codon. It was found that the carrier members were unaffected while the affected ones were homozygotes. Both mutations were confirmed using Sangersequencing. Conclusion: Herein, two novel pathogenic mutations were reported in Leber congenital amaurosis. Thiswould be of importance in genetic diagnosis and consultation of patients affected with LCA.

Abstract:

Introduction: Lung cancer is the second most common cancer in the world. The COR-L105 cell line isused as one of the most popular lung cancer cell lines in various studies. Long non-coding RNAs(lncRNAs) are a group of important cellular factors that play a key role in many processes such as growth, differentiation, replication, transcription, translation. LncRNA LINC00511 is a transcriptional regulatorand has been reported to increase its expression in various cancers. The aim of this study was to knockoutthe LINC00511 gene using a CRISPR/Cas9 in COR-L105 cells and to investigate its effects on cancerprogression and apoptosis. Methods: In this experimental study, two sgRNAs were designed for LINC00511 gene and cloned into two CRISPR vectors, separately. Two sgRNAs vectors were transferred to COR-L105 cells usinglipofectamine. After confirmation of LINC00511 gene knockout from target cells, changes in cellproliferation and apoptosis were assayed by MTT and flow cytometry. Furthermore, the expression ofgenes associated with apoptosis and tumorigenesis was examined by real-time PCR. Results: LINC00511 gene knockout from COR-L105 cell line was performed. The expression of BCL2, survivin, EZH2, c-MYC, and MAX genes showed a meaningful decrease related to the expression in treated cells (edited) compared to the control cells (p˂ 0. 05). Increased expression of p21 and p57 geneswas also seen in the edited cells (p˂ 0. 05). Decreased cell proliferation and increased apoptosis wereobserved in manipulated cells. Conclusion: Knocking of LINC00511 gene in lung cancer cell line caused apoptosis and decreased cellproliferation. Therefore, inhibiting the expression of the LINC00511 gene in cancer cells leads to thecontrol their proliferation.

Abstract:

Introduction: Family functioning can be one of the most important determinants of mental health of prisoners. Despite the research that has been done on the relationship between family functioning and mental health, few studies have examined the relationship between these two variables among prisoners. The aim of this study was to investigate the relationship between family functioning and mental health of prisoners on the verge of release in Kerman and Yazd prisons in 2019. Materials and Methods: The study was a cross-sectional one, and the data collection tools were Family Functioning Questionnaire (FAD) and General Health Questionnaire (GHQ). The statistical population was all 743 prisoners on the verge of release in prisons in Kerman and Yazd provinces. The results were analyzed using SPSS 23 software and Pearson's correlation and linear regression. Results: Out of prisoners, %95 were male and mean age was 27/9 ± 15/37. In terms of marital status, 522 (70. 3%) were married. Also, 378 (51%) of them had no history of imprisonment and 365 (49%) had a history of imprisonment of one to thirteen years. In terms of the type of crime committed, 335 (45%) of them committed drug-related crimes, 198 (27%) committed theft, and the rest (28%) committed other crimes. There was a positive and significant relationship between family performance and mental health of prisoners (p=0. 001). The regression results also showed that %13 of the changes in the mental health of prisoners were a function of their family functioning. Also, among the dimensions of family functioning, three dimensions of problem solving (β =0. 24), emotional intercourse (β =0. 13), and behavioral control (β =13. 13) were the strongest predictors of mental health of prisoners. Conclusion: Family functioning components had a positive and significant relationship with all aspects of mental health except social inefficiency. The lower the level of family functioning, the lower the mental health of family members.

Abstract:

Introduction: Cardiovascular disease is the leading cause of death in today's society, the main causes of which are a sedentary lifestyle and the use of high-fat and unbalanced diets. The aim of this study was to determine the effect of 4 weeks of aerobic exercise with octapamine supplementation on NLRP-3 gene expression and TRK-β protein synthesis in the heart muscle of rats fed with deep-fried oil. Materials and Methods: 30 male Wistar rats weighing 200-250 were randomly divided into 5 groups of 6: health control (CO), deep-fried oil (I), deep-fried oil + aerobic exercise (H), deep-fried oil + octapamine, supplementation (B), and deep-fried oil + octapamine supplementation + aerobic exercise (C). During the research period, 10 ml/kg deep-fried oil was given to the rats via gavage for 4 weeks. 81 mol/kg octapamine was intraperitoneally injected to the supplement groups. The rats in the exercise group also went through moderate intensity training in the first week (50% vo2max) and last week (65% vo2max) with treadmill. One way analysis of variance was used to analyze the data and Bonferroni test was used to determine the difference, using Graph pad prism version 8. Results: NLRP-3 gene expression was significantly reduced in the exercise group compared to the control group who did not receive any supplements and exercise (p<0. 05). Octapamine supplementation also decreased NLRP-3 (p<0. 05). The combination of exercise and supplementation significantly reduced NLRP-3 gene expression compared with the control group (p<0. 05). Changes in the amount of TRK-β protein in the exercise group did not show a significant difference compared to the control group that did not receive any supplements and exercise (p>0. 05). In the group that also received octapamine supplementation, compared to the control group, TRK-β values did not change and did not show a significant difference (p>0. 05). Combining exercise and supplementation significantly increased TRK-β gene expression compared to the exercise group (p<0. 05). Conclusion: Aerobic exercise training with octapamine supplementation may reduce the destructive effects of deep-heated oils on the expression of the gene associated with myocardial degradation and improve remodeling in myocytes.
