BIOLISTIC TRANSFORMATION OF WHEAT (TRITICUM AESTIVUM) USING CHITINASE AND GLUCANASE GENES

Abstract

 production of TRANSGENIC WHEAT (Triticum aestivum) was studied by CHITINASE and GLUCANASE genes using BIOLISTIC METHOD and pBI121 plasmid containing neomycin phospho transferase selectable marker gene under control of Nos promoter. Immature embryo explants from five genotype (arta, mogan, sisun, gascogen) cultivars and line A, excised from seeds and were placed on without antibiotic callus induction medium to produce embryonic calluses. Embryonic calluses, were bombarded with plasmids containing CHITINASE and GLUCANASE coated by gold nanoparticles. Then they were transfered to selective callus induction medium supplemented with 25 mg/l KANAMYCIN, and they were transferred to regeneration medium with 25 mg/l KANAMYCIN in order to producing shoots and roots. The maximum percent of transformation was realaited to line A with 4.8% and the minimum of them was realaited to sisun, gascogen and arta that there was not any transgenic plant. PCR and Dot Blot analysis showed that the putative transgenic plants consist at least one copy of either, CHITINASE, GLUCANASE and neomycin phospho transferase genes in their genome in comparison with control plants.
