Abstract

tuberculin is consisting of purified protein derivatives from tubercle bacilli. This product is injected to suspicious cases for diagnosis tuberculosis infection. In Iran Razi institute is an exclusive producer of bovine TUBERCULIN. Since 2004 Razi used AN5 strain to produce this product with other parts of the world.To this purpose genomic study of strain is inevitable. First these strain is inoculated on Lowenstein-Jensen medium. DNA required, is extracted from visible colony according to Van Soolingen protocol. To prove that these strain used in razi institute is belonging to Mycobacterium tuberculosis complex, PCR was done by specific primer INS-1 & INS-2 that is indicated IS6110 sequence. DNA is then digested by restriction enzyme PvuII & AluI. For a more detailed review after RFLP, southern blotting was performed. The results of IS6110_PCR showed 245bp region that is indicated of belonging these strain to Mycobacterium tuberculosis complex. Also enzymatic digestion by AluI & PvuII enzymes and HYBRIDIZATION BY PGRS PROBE provided genomic pattern for review and comparison with other strains used in the production of bovine TUBERCULIN in elsewhere.
