GENETIC DIVERSITY IN POPULATIONS OF FUSARIUM OXYSPORUM F.SP. CUMINI, THE CAUSAL AGENT OF CUMLNI WILT IN KHORASAN USING VEGETATIVE COMPATIBILITY GROUPS

Abstract

 Fifty isolates of FUSARIUM oxysporum were recovered from vessel tissues of crown and roots of CUMIN plants collected from major producing areas in Khorasan province during 1999-2000 growing seasons using Nash - Snyder medium. Forty eight isolates pathogenic causing wilt in CUMIN and identified as F. oxysporum f. sp. eumini.Four hundred and eighty three nitrate non utilizing (nit) mutants were generated from 50 isolates of F. oxysporum on three media, potato dextrose, minimal and Czapek agar amended with 1.8%, 2.5% and 3% potassium chlorate.Nit mutants were divided into three phenotypic classes (nit 1, nit 3 and Nit M) based on their growth on media containing different nitrogen sources (sodium nitrate, sodium nitrite, hypoxanthin, ammonium tartrate). Of the NIT MUTANTs obtained from F. oxysporum isolates, 74.5%, 12% and 13.5% were nit 1, nit 3 and nit M, respectively. NIT MUTANTs were used to force HETEROKARYON and to determine distribution of VCGs and their relation to pathogenicity and geographic origin. Six VCGs were recovered for F. oxysporum isolates. Two of these belonged to the nonpathogenic isolates, and the remaining four belonged to the Foc populations. VCGs A comprised 44 isolates (93/6% of the population) that had been collected from Ferdos city, while each of the remaining VCGs represented by a single isolate from different parts of the province. One isolate was self-incompatible. Based on these results specific relation was observed between VCGs and geographic origin of the isolates in F. oxysporum f. sp. CUMINi. It seems that, VCG was able to differentiate pathogenic and non pathogenic strains.
