Abstract

 Damage and tear of ARTICULAR CARTILAGE that are due to aging and over loading are inevitable. There are some different methods in the literature that are used for healing and repairing of cartilage. One of the latest methods is TISSUE ENGINEERING. In this method, a BIODEGRADABLE porous scaffold with cells is used for tissue repair. In TISSUE ENGINEERING, by changing the physical factors such as pH, oxygen tension, osmolarity and hydrostatic pressure, the cell metabolism can be stimulated on a suitable scaffold. One of the suitable scaffolds for ARTICULAR CARTILAGE TISSUE ENGINEERING is POLYURETHANE that has a wide range of properties. Degrapol® is a type of commercial POLYURETHANEs that was used in this research. In this research, the effect of pH (7.4, 6.5), osmolarity (280, 380,480 osm/L), oxygen tension (1%, 5%, 21%) and hydrostatic pressure at 4MPa on chondrocyte metabolism on Degrapol, was evaluated. The results showed that these factors could significantly affect cell metabolism. The best conditions was obtained at pH=7.4, 5% O2, osmolority=380 osm/L under hydrostatic pressure. The metabolism conditions were evaluated by measuring the cell attachment after 2 h, cell viability after 3 days, lactate production in day 1, 2 and 3 and GAG (glycosaminoglycan) production in day 3, 7 and 14. The final results show that Degrapol can be a suitable scaffold for chondrocyte metabolism and is useful for ARTICULAR CARTILAGE TISSUE ENGINEERING.
