Abstract

 In this study OPTIMIZATION of medium nutrients and culture conditions for improvement of CATALASE production by CORYNEBACTERIUM GLUTAMICUM PTCC 1532 were performed using one-factor- at-a-time and orthogonal array designs. The one-factor-at-a-time method was used to study the effects of various carbon and nitrogen sources, temperature and initial pH on CATALASE production. The results showed that, the optimal temperature and initial pH for CATALASE production were 40 ْ C and 8.5, respectively. The optimal concentration of nutritional components for improved CATALASE production after Taguchi ’s method were determined as 4g l-1 glucose,1g l-1 Bactopepton, 0.2 g l-1 Na3- Citrate, 0.2 g l-1   KCl , 0.3 g l-1   Na2Co3 , 0.1 g l-1   MgSo4.7H2O, 0.00025 g l-1  Mncl2.4H2O, 0.005 FeSo4. The experimental results showed that CATALASE production could be improved by varying the composition of MEDIUM COMPONENTS. The obtained results showed that maximum CATALASE production were in basal and optimized medium after Taguchi method were 3.33 and 36.15 (U/ml) about 11-fold higher than the basal medium.
