BIOCONTROL OF TRANSFORMANT TRICHODERMA HARZIANUM RESISTANT TO BENZIMIDAZOLE AND CALCULATION ITS EC50

Abstract

 Recently a method use for identifying the TRANSFORMATION of resistance gene of BENZIMIDAZOLE into TRICHODERMA HARZIANUM for BIOCONTROL effect. This resistance level of the transformants is higher than the chemical dose used in plant disease control in the field. Therefore, it is safe to use those transformants in an integrated plant disease management program with the chemicals. In this study, BENZIMIDAZOLE resistance gene in the transformants was very resistant to BENZIMIDAZOLE and the isolates with BENZIMIDAZOLE resistance of TRICHODERMA HARZIANUM were achieved.To this way, first transformants cultured on the Potato Dextro Agar (PDA) that was used for growing the fungi (control growth) and measure total growth after 5 days, then transformants cultured on the PDA medium contain different concentration of BENZIMIDAZOLE: 0, 0.1, 1, 10, 100, 1000 mg/ml (treatment growth) and measure total growth during 3 weeks. The inhibition growths of the fungi were calculated out as follows:Inhibition growth=1- (Treatment growth/Control growth).The resistance level of those transformants is as high as 1000 mg/ml. In this research we investigate resistance level and stability of the resistance for the transformants isolate of TRICHODERMA HARZIANUM. For detection of resistance stability, transformants were subcultured on an unselective medium for 10 times. Then those were tested on PDA with different concentrations of BENZIMIDAZOLE. The results shows every one of transformants can survive on medium contain PDA and BENZIMIDAZOLE with concentration more than 1000 mg/ml and this genetic resistance is stabled.As also, we calculate EC 50. That is effective concentration of carbendazim that can survive 50% of TRICHODERMA HARZIANUM in that concentration (EC 50 average about 1200 mg/ml).This study show transformants of TRICHODERMA HARZIANUM are effective BIOCONTROL for application in plant disease.
