DESIGNING A PCR INTERNAL CONTROL FOR DIAGNOSIS OF FREE LIVING AMOEBA

Abstract

 Free-livings Amoebas were diagnosed first time in 1958. They target CNS and other human organs (like eyes) so cause death or permanent paralysis. The miss diagnoses of these parasites lead to some problems. The aim of this study was to prepare an INTERNAL CONTROL for using in PCR reaction for diagnosis of free living amoeba. For this reason, the 1056 bp of 18S rDNA of achantamoeba genome was amplified and cloned into a cloning vector plasmid. Recombinant plasmid was digested by PpmuI restriction enzyme and relegated again added with 300 bp DNA fragment. While done a PCR reaction for sample diagnosis with this recombinant plasmid, 1356 bp will appear whether it is positive or negative. The results of this project are offering in form of diagnostic kit.
