INVESTIGATION ON FREQUENCY OF LACTOBACILLUS SPP. BACTERIA IN DUODENUM, JEJUNUM, ILEUM AND CECUM OF POULTRY USING DENSITOMETRY TECHNIQUE

Abstract

LACTOBACILLUS SPP. have critical role in prevention of some pathogens bacteria colonization e, g. Salmonella spp. and Clostridium spp. in gastrointestinal tract's POULTRY. These bacteria increase POULTRY resistance against diseases and improve their immune system and performance. In order to determining of frequency of LACTOBACILLUS SPP. in duodenum, jejunum, ileum and cecum of broiler chickens, it was used DENSITOMETRY technique based on PCR results. Digestive tract contents were removed and extracted their DNA. It was obtained specific bands for detecting of LACTOBACILLUS SPP. and all bacteria. It was determined relative population of LACTOBACILLUS SPP. relative to total GUT bacteria by means of DENSITOMETRY technique. Result analysis was showed LACTOBACILLUS SPP. consists 2.62% of total duodenum bacteria.Also it was showed LACTOBACILLUS SPP. consists 3.58% of total jejunum bacteria. Meanwhile LACTOBACILLUS SPP. consists 2.34% of total ileum bacteria.Furthermore it was showed LACTOBACILLUS SPP. consists 2.93% of total cecum bacteria.Relative population of LACTOBACILLUS SPP. in lower segments i.e. ileum and cecum were as same as upper segments i.e. duodenum and jejunum. Meanwhile among different ages, the highest and lowest relative population of LACTOBACILLUS SPP. obtained in 14 and 30d of ages respectively. The highest relative population of LACTOBACILLUS SPP. obtained in jejunum of broilers in 4d of ages (5.97%).Also, the lowest relative population of LACTOBACILLUS SPP. obtained in jejunum of broilers in 30d of ages (0.88%).At 4d of ages, the highest relative population of LACTOBACILLUS SPP. obtained in jejunum.But, At 14 and 30d of ages, the highest relative population of LACTOBACILLUS SPP. obtained in ileum. From obtained results, it was showed LACTOBACILLUS SPP. populations are variable in various intestine segments and correlated with their functions and physicochemical environment.
