Effect of humic acid on germination indices and antioxidant enzymes of canola seed (Brassica napus L. ) under the influence of toxicity of nanoparticles of copper oxide

Abstract

 Concerns about Nanoparticles environmental pollution risk have been increasing globally due to increasing in production of Nanoparticles in recent years and their use in diverse cases such as medical, imaging, catalysts, semiconductors, cosmetics, microelectronics, various material coatings etc. The purpose of this experiment was to determine the effects of copper oxide nanoparticle on some antioxidant enzymes, protein and germination indices of canola seeds and also to study the humic acid effects on reducing of Nanoparticles toxicity in laboratory conditions. Then, an experiment was conducted to investigate the effects of copper oxide nanoparticle in 9 concentrations of 0, 250, 500, 750, 1000, 1250, 1500, 1750, 2000 mg. L-1 and humic acid in two concentrations of 0 and 100 mg. L-1 in a factorial experiment as completely randomized design with three replications. The results showed that increasing the concentration of copper oxide nanoparticle resulted in rising the activity of all four enzymes i. e. superoxide dismutase, ascorbate peroxidase, catalase, and peroxidase 5, 2. 19, 2 and 2. 35 times respectively, and decreasing the protein content twice as much. Also, germination indices including radicle length, plumule length were decreased 39. 00 and 6. 56 time, respectively. Humic acid application increasing the toxicity effects of copper oxide Nanoparticles, especially at 2000 mg. L-1, so that the radical and plumule length, radical and plumule fresh weight were 9. 68 and 3. 0, 6. 0 and 5. 50 times than of the control respectively. It can be stated that humic acid can increase the activity of antioxidant enzymes and increase plant growth by increasing pseudo-hormonal activity.
