Abstract: 

Background and Objectives In recent years, Natural auto Abs (NAAbs) are considered as key factors in maintaining the immune homeostasis; NAAbs are also known as biomarkers in various diseases. This research, for the first time, was performed to identify NAAbs that are produced against RBCs following some physiologic changes in patients in comparison to healthy blood donors.Materials and Methods In this fundamental-applied research, two groups of samples including 600 healthy blood donors and 324 patients who have shown pathologic disorders participated. All samples were screened for NAAbs but the identification panel was performed only in the case of positive reaction. Results were analyzed through binominal test and by SPSS 16.Results NAAbs were detected in 8.6% of healthy population and 17.9% of the patient population, all determined as positive reactions. Only two cases (1 anti-A1 and1 anti-P1) out of 30 were identifiable among the healthy group with auto antibody; however, in the patient group with auto antibody, 19 anti-IH cases, 1 anti-H cases, 2 anti-I cases, 2 anti-P1 cases, and 6 inconclusive cases were identified.Conclusions A significant difference between the frequency of NAAbs reactions against RBCs in the two groups was observed (p=0.03). Furthermore, anti-IH increases in hematologic diseases.

Abstract: 

Background and Objectives To the opinion of psychologists, job satisfaction has been defined as the action of coordination between individual needs and professional values. Therefore, evaluating the rate of job satisfaction is one the concerns of the professionals. The objective of this survey was to assess the rate of job satisfaction among the staff of IBTO headquarters (HQ) and Tehran Blood Transfusion Center (TBC).Materials and Methods The study is a descriptive-analytical and cross- sectional study which was done voluntarily on 120 staff of the IBTO HQ and 94 staff of TBC. To gather the data, JDI questionnaire was used.The data was analyzed by MANOVA test. P<0.01 was identified as the criteria for significant differences.Results The average rate of job satisfaction in IBTO headquarters was measured as standard deviation.This rate was 21.32 in Tehran Blood Transfusion Center. The results indicate the higher job satisfaction of the staff of TBC compared to IBTO HQ. The highest unsatisfactory variables pertained to job promotion, supervisors and the colleague relationship.Conclusions Job satisfaction is an indicator for the performance improvement of the staff leading to success of an organization. While the highest unsatisfactory variables pertained to job promotion, supervisors and the colleague relationship, the necessary interventions have to be made in order to clarify and revise the ways of job promotion and to establish a competent intersectorial relationship between the staff.

Abstract: 

Background and Objectives: Many different factors are effective in the safety of blood donations. One of the main factors is the education status of blood donors which is the focus of the present research.Materials and Methods: Our study is based on a cross-sectional method which was done for all donations during 2007-2008 (3519254 blood donations). The necessary data such as educational degree, sex, age, history of previous donation and the confirmed positive results (Ab-Ag HIV HCV Ab and HBsAg) were collected from the database of e.projesa & negareh blood transfusion software.(p<0.05).Results: Among 3319254 blood donations, 7% were female and 93% were male. The donors with the academic degree showed to be HIV positive in 1.9 units per 100000 blood donations, 3.7 for donors with diploma and 7.5 for the illiterate (p<0.001). The rates for HCV positivity were 0.4 for blood donors with the academic degree, 0.9 for the ones with diploma, and 1.8 units per 1000 for the illiterate (p<0.001). For HBS Ag positivity the figures were 2.2 for blood donors with the academic degree, 2.8 for those with diploma, and 5.8 and 5.6 units per 1000 for the illiterate (p<0.001).Conclusions: These findings showed that the level of education has a direct correlation with blood safety.The results can help Iranian Blood Transfusion Organization adopt the best strategies in recruiting the eligible blood donors from the low risk communities.

Abstract: 

Background and Objectives: Among the key issues in the blood supply chain design, the location of blood collection centers is essential to ensure the easy access of donors for blood donation and distribution of blood products to hospitals. The aim of this study was to calculate the optimal number and location of blood collection and processing centers, blood collection centers and mobile teams based on clinical demand, cost of construction and operating cost of these centers in East Azerbaijan province.Materials and Methods: The study was a mathematical calculation modeling on the population of East Azerbaijan province. Three six-month periods with the first and the second in 1393 and the third in 1394 were used for calculating cost. GAMZ mathematical model was implemented and the results were extracted.Results: The results of the GAMZ application showed that a suitable locations for the establishment of a blood collection and processing center were in Maragheh and Mianeh, and the optimal locations for blood collection center were in Tabriz and Ahar. What is more, one mobile team could move among Marand, Tabriz, Ajabshir, Bonab, Malekan, Maragheh and Hashtrood cities in alternation with the cost of transport to be reduced.Conclusions: The optimal determination of number and locations of blood collection and processing centers, blood collection centers and mobile team decreased the cost of blood collection services. In future, to plan for any changes in order to increase or decrease the number of blood centers in all provinces, the mathematical modeling in other conditions should be considered.

Abstract: 

Background and Objectives: Acute lymphoblastic leukemia (ALL) is the most common malignancy among children. The use of natural compounds has invoked a lot of interest in treatment of malignancies due to the adverse effects of chemotherapy treatment. Artemisia annua was reported to show cytotoxic effects on various cancer cell lines. In this study, cytotoxic effects of Artemisia annua extract were investigated on acute lymphoblastic leukemia cell lines.Materials and Methods: In this basic research, Nalm-6 and Reh cells were cultured and then treated with various concentrations of Artemisia annua extract; afterwards, the cell viability was evaluated using MTT assay for 48 and 72 h. The Caspase 3 activity assay and flow cytometry following Annexin V and Propidium iodide staining were used to assess apoptosis. Statistical analysis was evaluated by one-way ANOVA test.Results: A. annua extract showed IC50 of 90 mg/ml on Nalm-6 and IC50 of 70 μg/ml on Reh cells after 48 h; the cytotoxic effect after 72 h was noticeable (p<0.001). Flow cytometry results also show that A. annua extract concentration of 40 mg/ml increases the percentage of apoptotic cells compared with control groups (p<0.05). Significant increase in Caspase 3 activity was observed after treatment with Artemisia annua compared with control groups (p<0.01).Conclusions: Our results show that methanolic extract of aerial parts of A. annua exerting cytotoxic and inhibitory effects on Nalm-6 and Reh cells.

Abstract: 

Background and Objectives The frequency of deregulated PI3K in acute lymphoblastic leukemia (ALL) coupled with the critical role of this signaling pathway in the acquisition of chemo-resistant phenotype lend compelling weight to the application of PI3K inhibitors for the treatment of ALL. In this study we aimed to evaluate the effect of selective p110d inhibitor, GS-1101 on acute lymphoblastic leukemia Nalm-6 cells.Materials and Methods Nalm-6 cells were treated with different concentrations of GS-1101. In order to determine the cytotoxic and anti-proliferative effects of the drug, MTT and trypan blue exclusion assays were performed, respectively. Afterwards the effect of GS-1101 on cell cycle progression was evaluated using flowcytometry. Finally, Rq-PCR was applied to evaluate the mRNA expression level of cell cycle- and apoptotic-related genes in GS-1101-treated cells.Results: Our results showed that GS-1101 not only reduced the number of viable cells but also hampered the metabolic activity of inhibitor-treated Nalm-6 cells both in dose- and time dependent manner. Moreover, we found that the anti-proliferative effect of GS-1101 is mediated, at least partially, through the induction of G1 arrest as a result of up-regulated p21 expression level and accumulation of cells in sub-G1 phase of cell cycle. The results of Rq-PCR also demonstrated that GS-1101 exerts an inductive effect on the mRNA expression level of Bax, as the most important pro-apoptotic gene.Conclusions: p110d isoform inhibitor, GS-1101 shows both apoptotic and anti-proliferative effect on Nalm-6 cells through inducing cell-cycle arrest via up-regulation of cycline-dependent kinase inhibitor, p21 and upregulation of pro-apoptotic-related gene.

Abstract: 

Background and Objectives: Stem cell factor (SCF) is a 28-40 kDa glycoprotein which plays an important role for the proliferation and differentiation of hematopoietic stem cells (HSCs). SCF binds to the C-kit receptor, and improves the survival of HSCs in vitro. SCF is one of the essential supplements for cultivation of HSCs in vitro. It plays a vital role to increase the number and size of HSC colonies.Materials and Methods: In this experimental study, glycosylation of SCF does not seem to influence its biological activity; therefore, it would be possible to produce it in prokaryotic expression system. The aim of this study was isolation, cloning and expression of SCF in the Rosetta expression host In addition to the features of prokaryotic expression system, Rosetta was expected to provide rare codons of eukaryotic proteins and increase the recombinant protein expression level.Results: The SCF coding sequence was isolated and amplified using the specific primers and cloned in E.coli TOP10 using pET-32a expression vector. The recombinant construct was confirmed by PCR, digestion and sequencing. The recombinant vector was transformed to the expression host, Rosetta.Conclusions: The SCF coding sequence was successfully isolated and cloned into the expression vector pET- 32a, followed by recombinant protein expression in Rosetta host strain.

Abstract: 

Background and Objectives: Stomach cancer is currently ranked as the fourth most common cancer and the second regarding to the mortality rate. Environmental and genetic factors both play crucial role in the onset of inflammation in the stomach and progression of the cancer. In a majority of gastric cancer cases, Helicobacter Pylori infection is a responsible factor. Nowadays, the micro RNAs are pinpointed as the powerful molecules in the field of disease genetics. In the present study by implementing several bioinformatics database, we have undertaken a research on the association of miR-146a with the expression of the genes involved in cell signaling pathways in gastric cancer development.Materials and Methods: This type of study was theoretical bioinformatics. The first step to candidate the most important miRNAs is to collect data from the previous studies; then, miRWalk 2.0 and miRTarBase databases were chosen for primary studies. The final review was made at David database in order to achieve pathways involved in gastric cancer.Results: Based on the results from this study, miR-146a was predicated as a key factor in preventing the cancer progression by affecting an increase on metastasis and tissue attack and preventing apoptotic pathways.Conclusions: Data revealed that the expression of miR-146a increased the progress of gastric cancer. Thus, the analysis of the miR-146a expression could be accounted as a valuable marker for prognosis of gastric cancer.
