AN INVESTIGATION OF ANTHOCYANIN PRODUCTION’S ABILITY BY RHIZOBIUM SP. IN CULTURE MEDIUM AND SYMBIOSIS FORMS

Abstract

ANTHOCYANINs are a group of a flavonoids pigments vastly existing in the nature.  The positive impression of these pigments on the health of human is reported in some researches. ANTHOCYANINs are famed for their anti-oxidation, anticancer, anti-phlogistic. In regard to the importance of bio-pigments, the present thesis is aimed to study the production ability of this pigment in RHIZOBIUM SP.In this cause, sampling from Lucerne and Haricot Roots were made in Karaj, Khomein and Mashad Zones. After maroscopic and microscopic examining of the NODULES, existence of ANTHOCYANINs was studies in both condition, symbiosis and culture medium. Pigments were extracted by using methanol acid solvents in symbiosis condition and acetone-methanol in the culture medium condition. Then, the extracted pigments underwent analysis by implementation of thin layers chromatography and gas chromatography-mass spectrometry. For optimization of pigment production in YMA base cultural medium, the impression of four Carbon Base (glucose, sucrose, lactose and mannitol) three organic and inorganic nitrogen base (potassium nitrate, ammonium chloride, and fertilizer extraction); three temperatures 25, 30 and 35 centigrade and six different pH (5, 6, 5, 6, 7 and 8 underwent studies).The results in this research revealed that the extracted pigments in ANTHOCYANINs symbiosis condition and its derivations and extracted pigments in CAROTENOID PIGMENTal cultural medium conditions are of astaxanthin class. Mannitol sugar showed an additional impression on crowd bacteria among carbon resources and fertilizer extraction. Also, the most suitable pH 7 and 30oC degree temperature were determined. In gas chromatography method, in addition to ANTHOCYANINs pigments, some vaporizable compounds seen in nodular extraction specimen along with acidic methanol solvent is reported.
