STUDY ON EFFECT OF GENOTYPE AND EXPLANT TYPE ON SHOOT REGENERATION IN RAPESEED (BRASSICA NAPUS L.)

Abstract

RAPESEED is one of the most important oil crops in the world. Successful gene transfers to plants are depends on the development of efficient transformation and SHOOT REGENERATION methods. The aims of this research are to evaluate the ability of different  RAPESEED GENOTYPE for regeneration in in-vitro condition. Here, the seven commercially important cultivars of B. napus (PF-7045-91, SLM-046, Westar, Global, Ceres, Option500 and Hyola 401) were compared for SHOOT REGENERATION in a standard condition. The Seeds of these cultivars were surface sterilized and planted on the germination medium. After 5 days the COTYLEDONs were excised in such a way that they included 2 mm petiole at the base. Care was taken to eliminate the apical meristem which sometimes adheres to petioles. In order to develop the shoot, the excised COTYLEDONs were placed on induction medium and the petioles were embedded in the solid medium to a depth of approximately 2 mm. The 21-day-old COTYLEDONs were cultured on callus and subsequently on shoot induction medium. The explants were maintained on shoot induction medium under standard light and temperature conditions. During the incubation periods many shoots appeared on over cut end of the COTYLEDONs or on induced callus in HYPOCOTYLS. The shoots were counted and transferred to elongation and rooting medium. Results showed that there is a significant difference in SHOOT REGENERATION potential between cultivars and explants. The highest regeneration frequency from COTYLEDONs and HYPOCOTYLS was achieved at PF-7045-91 cv (84.7%) and SLM-046 cv (9.2%) respectively. Generally the COTYLEDONs were more regenerable than HYPOCOTYLS. Our study shows that seedling COTYLEDONs of commercial cultivars of RAPESEED are amenable to multiple shoot formation with high regeneration frequencies, and could be used for transformation experiments.
