EFFECTS OF CAFFEINE AND IRON ON LIPID PEROXIDATION, ACROSOME REACTION AND DNA INTEGRITY OF BULL SPERMATOZOA IN THE PRESENCE OF TAURINE AND ALBUMIN

Abstract

 The effects of environmental agents on fertility are a major concern in reproductive physiology. Accumulation of these toxic agents in animal’s body is the most important factor in infertility. Therefore, evaluation of respective impacts on the normal sperm function is imperative. In the present study, semen samples were obtained freshly from Holstein bulls. The lipid PEROXIDATION (LPO) was evaluated in terms of thiobarbituric acid (TBA)-reactive substances (TBARS), particularly malondialdehyde (MDA). The amount of TBA+MDA complex was measured in the presence of LPO promoter (ferrous+ascorbate), taurine (0.5-5 millimolar), and albumin (0.25-5.25%). The next step was addition of different concentrations of CAFFEINE and IRON (200-1000 micro M) with/without antioxidant supplementation to the BULL SPERM samples as compared to controls. All treatments showed a significant increase in the MDA level/LPO rate. Following addition of antioxidants to media, a reduction in the generation of MDA occurred. In the treated groups, no halo was found in gelatin slides indicating the impairment in the sperm membranes after application of CAFFEINE and IRON. The extent of DNA breaks (by comet assay technique) in treated sperm samples was evaluated with silver nitrate staining. We found a slight increase (p>0.05) in the number of spermatozoa with DNA damage as compared to controls.
