EFFECT OF ESTROGEN ON ACQUISITION OF MORPHINE–INDUCED CONDITIONED PLACE PREFERENCE USING BRAIN MICRODIALYSIS IN ADULT MALE RAT

Abstract

 Tolerance and dependence are the two main issues of drug abuse including morphine. It has been suggested that many substances are involved in morphine-induced psychological dependence. The current study aimed to determine whether or not long-term intracerebroventricular (ICV) perfusion of estrogen could modify the acquisition of morphine-induced conditioned place preference (CPP) in adult rats. Male Wistar rats weighing 250-300g were anesthetized with chloral hydrate (500mg/Kg, ip). Guide cannula was implanted in the right lateral ventricle using stereotaxic instrument. After 1 week of recovery from surgery, rats received morphine subcutaneously and long-term ICV perfusion of estrogen (2 μL/min for 2 hours) using a microdialysis probe and the guide cannula. The biased method was used to determine the induction of CPP in rats. Changes were monitored in all groups, and statistical analysis was carried out using absolute data and one-way ANOVA. The results indicate that subcutaneous administration of different doses of morphine sulfate (0.5-10 mg/Kg) produced a dose-dependent CPP in all experimental groups. The ICV perfusion of estrogen (2mg/Kg) did not produce reliable CPP; however, long-term ICV perfusion of estrogen significantly increased the acquisition of morphine-induced (10mg/Kg) CPP. These data suggest that estrogen influences morphine-induced euohpria and/or reward.
