FRUIT FLY AS A MODEL FOR PAIN STUDIES

Abstract

 Pain research using animal models is related to ethical concerns, so invertebrates and insects have been recommended by researchers and animal ethics organizations. It appears that DROSOPHILA model for screening of putative analgesics would be advantageous in discovery of physiopathology of PAIN and pharmacology of new drugs. In this research, DROSOPHILA LARVAE were employed to be suggested as a good replacement of vertebrates. For this purpose, 3rd stage of d. Melanogaster LARVAE was used. Thermal nociceptive reactions (threshold and maximum) were recorded by hot plate assay in temperatures between 35°с-47°с. Writing test also was conducted in various concentration of acetic acid (%10-%100) and rolling movement responses was recorded. These tests were done in each concentration and temperature in separate groups (n=7). MORPHINE at different concentrations was administrated by floating the LARVAEs in the MORPHINE solution. First response (threshold) at 40°с and maximum response at 47°с (p<0/001) were recorded in hot plate test. By increasing the acetic acid fluid concentration (each step for %10), there was a significant attenuation of animals response and the maximum was recorded at the concentration of %40 (p<0/001). MORPHINE diminished responses to noxious stimulus in this suggested animal model of PAIN research (p<000.1). According to our results, it can be suggested that DROSOPHILA melanogaster LARVAE may be used as a model for PAIN assessments as well as drugs which are used as anti-nociceptives or hyperalgesic factors.
