MORPHINE ORAL CONSUMPTION REDUCED VISUAL SYSTEM DEVELOPMENT IN RAT EMBRYOS

Abstract

 Among an important systems in life time is VISUAL SYSTEM. The possible defects resulted from oral MORPHINE consumption in female pregnant Wistar RATs on their embryos were investigated in the present study. Pregnant female RATs were divided into two groups and studied. Experimental group receive MORPHINE (0.5 mg/ml) solution instead of water during the experiment were as the control group receive tap water. Blood samples were taken from RATs’ retro-orbital sinus for blood CORTICOESTRONE concentration on 13th embryonic day. On the 17th embryonic day (E17), the pregnant RATs were killed by cloroform and their embryos were taken out surgically. The embryos were then fixed in formalin 10% solution for 30 days. Then the embryos were physically examined and their lengths and weights were determined by a clipper and digital scale respectively. Then the embryos were sectioned in their orbital area and stained by Hematoxiline and Eosin method. The stained slides were evaluated for any morphological changes by MOTIC software and light microscope. Results have shown that plasma CORTICOESTRONE level was higher in the experimental group. The distance between the embryos eyes was lower in experimental group. From the microscopic view, General eye size was significantly reduced in experimental group. In addition, LENS sizes, CORNEAl, and RETINAl thicknesses (in fovea area) also were reduced in the experimental group, and induced is a delay in migration of LENS cells in experimental group. These results indicate that MORPHINE exposure can interrupt VISUAL SYSTEM development in the embryos but the role of CORTICOESTRONE also must be considered.
