Abstract

 THIS STUDY WAS DESIGNED TO INVESTIGATE THE ROLES OF LIGHT AND HEAVY CHAINS OF ANTI- DNA ANTIBODIES IN BINDING TO DNA. THE MAIN OBJECTIVE WAS TO PREDICT THE SEQUENCE OF THE EFFECTIVE REGIONS OF EACH CHAIN CONFERRING THE DNA SPECIFICITY OF THESE ANTIBODIES. TO DO THIS TASK, THE PROTEIN SEQUENCES OF DNA BINDING AND NON DNA BINDING ANTIBODIES WERE CODED USING THE PHYSICOCHEMICAL PROPERTIES OF THEIR AMINO ACIDS AND THE RESULTS WERE USED AS THE TRAINING PATTERNS OF A MULTI-LAYER PERCEPTRON NETWORK. FOR EACH CHAIN, A SEPARATE NETWORK WAS USED AND TRAINED BY THE BACK PROPAGATION METHOD. BY FOCUSING ON THE CDR REGIONS, PREDICTIONS WITH 70.4% AND 54.5% CORRECTNESS WERE OBTAINED FOR THE NETWORKS RELATED TO THE HEAVY CHAIN AND LIGHT CHAIN, RESPECTIVELY. THIS CONFIRMS THE EXPERIMENTAL RESULT THAT IN BINDING TO DNA, THE LIGHT CHAIN IS LESS IMPORTANT THAN THE HEAVY CHAIN. CONSIDERING THE OBTAINED RESULTS ABOUT THE LIGHT CHAIN, THE INVESTIGATIONS USING ARTIFICIAL NEURAL NETWORKS CAN FOCUS ON THE HEAVY CHAIN. BY SOME MODIFICATIONS IN THE NETWORK, INPUTS AND TRAINING, THE RESULTS REGARDING THE HEAVY CHAIN WERE IMPROVED IN THE CURRENT STUDY, IN COMPARISON TO SIMILAR PREVIOUS STUDIES.
