USING OF LASER SPECTROSCOPY FOR DETECTION OF AFLATOXIN IN PISTACHIOS NUTS

Abstract

 BACKGROUND: AFLATOXINS ARE MYCOTOXINS WHICH CAN BE FOUND IN A WIDE RANGE OF AGRICULTURAL PRODUCTS.IN THE PRESENT PAPER LASER SPECTROSCOPY IS USED TO INTRODUCE AN ACCURATE AND FAST AFS CONCENTRATION MEASUREMENT METHOD ON THE PISTACHIO SURFACE WITHOUT EXTRACTION AND ANY OTHER PREPARATION BEFORE THE MEASUREMENT PROCEDURE. THIS METHOD IS USED FOR MEASUREMENT OF DIFFERENT TYPE OF AFS IN PISTACHIO AND CAN BE EMPLOYED IN LINE IN INDUSTRIAL PREPROCESSING [1-4].METHODS: PISTACHIO NUTS WERE COLLECTED FROM RAFSANJAN ORCHARDS. SAMPLES WERE STORED IN 6 SEPARATE CONTAINERS, AN INTACT PISTACHIO CONTAINER IS KEPT IN A CLEAN, DRY AND COOL MEDIUM, THE PISTACHIO OF OTHER FIVE CONTAINER SULLIED BY A SUSPENSION SOLUTION OF TOXIN INCLUDING OF ASPERGILLUS FLAVUS AND ASPERGILLUS PARASITICUS. PISTACHIO NUT IS ILLUMINATED BY AN ULTRA VIOLET (UV) LASER IRRADIATION, AND THE SAMPLE FLUORESCENCE RADIATION IS ANALYZED BY AN ULTRA VIOLET-VISIBLE (UV-VIS) SPECTROMETER. SELECTION OF LASER WAVELENGTH IS BASED ON THE SAMPLE UV ABSORPTION SPECTRUM WHILE SPECTROMETER BAND WIDTH MUST BE SELECTED ON THE FLUORESCENCE EMISSION SPECTRUM.RESULTS: OUR MEASUREMENTS SHOW THAT LASER SPECTROSCOPY HAS THE ABILITY FOR THE ON LINE AFS CONCENTRATION MEASUREMENT IN THE PISTACHIO COLLECTION PROCESSES.CONCLUSION: LASER FLUORESCENCE SPECTROSCOPY IS A POWER FULL METHOD FOR CONCENTRATION MEASUREMENT OF ALL AFLATOXIN KINDS BOTH IN LIQUID AND SOLID MEDIA. THE RESULT OF PISTACHIO MEASUREMENTS ARE COMPARED WITH THOSE ARE MEASURED BY HPLC METHOD. OUR MEASUREMENTS SHOW THAT LFS HAS THE ABILITY FOR THE ON LINE AFS CONCENTRATION MEASUREMENT IN THE PISTACHIO COLLECTION PROCESSES.
