PRODUCTION OF ALKALINE PROTEASE BY BACILLUS THURINGIENSIS H14

Abstract

 ALKALINE PROTEASE ACTIVITY DURING BATCH CULTURE OF BACILLUS THURINGIENSIS H14 WAS EVUATED IN DIFFERENT PHYSIOLOGICAL PHASE. THE EXPERIMENTS WERE CONDUCTED IN SHAKEING FLASKS USING SEVERAL SYNTHETIC MEDIA CONTAINING GLUCOSEAS CARBON SOURCE THE CULTIVATION LASTED 96HIN THE TEMPERATURE WAS CONTROLED AT 30O. THE EFFECTS OF NITROGEN SOURCE AND AGITATION RATE WERE EXAMINED IN ORDER TO FIND THE OPTIMUM CONDITIONS. THE RESULTS SHOWED THAT, MAXIMUM PROTEASE ACTIVITY OCCUR AT THE DEATH PHASE OF THIS BACTERIUM AND AT START OF SPORULATION PHASE. THE MAXIMUM ALKALINE PROTEASE ACTIVITY WAS OBTAINED USING YEAST EXTRACT AS NITROGEN SOURCE AT 350 RPM AFTER 27 HOURS AT 11.76 U/ML. THUS YEAST EXTRACT WAS FOUND BETTER SOURCE AND 350 RPM IS BEST AGITATION RATE FOR ENZYME PRODUCTION OF THIS STRAIN.
