ENCAPSULATION OF PROTEIN DRUG WITH PLGA BY ELECTROHYDRODYNAMIC SPRAYING

Abstract

 ENCAPSULATED PHARMACEUTICAL MATERIALS HAVE RECEVIED MUCH ATTENTION FOR MANY YEARS DUE TO THEIR INCREASED THERAPEUTIC EFFICIENCY, BIOAVAILABILITY AND HIGH DISSOLUTION RATE. THERE ARE DIFFERENT PROCEDURES FOR ENCAPSULATING AND THE CHOICE OF PROCEDURE INFLUENCES THE SIZE OF PARTICLES FOR INTENDED APPLICATIONS. ELECTROHYDRODYNAMIC SPRAYING IS USE AS A NOVEL METHOD TO PRODUCE DRUG NANOCAPSULES.THE AIM OF THIS STUDY IS TO PROVE THE ENCAPSULATION OF PROTEIN DRUG INSIDE BIODEGRADABLE POLYMERIC NANOCAPSULES BY ELECTROSPRAYING. BOVINE SERUM ALBUMIN(BSA)WAS CHOSEN AS A MODEL PROTEIN DRUG.THE EFFECTS OF SYSTEM OPERATION PARAMETERS AND FORMULATION OF SPRAYING SOLUTION WERE TESTED.MORPHOLOGY AND DIAMETER OF NANO-CAPSULES WERE INVESTIGATED USING SCANNING ELECTRON MICROSCOPE(SEM).SMALLER NANOCAPSULES CAN BE GENERATED BY DECREASING THE BSA CONCENTRATION OR SPRAYING LIQUID FLOW RATES AND INCREASING THE VOLTAGE. THE NANOCAPSULES PRODUCED WERE IN THE SIZE RANGE 85-260 NM. THERMAL ANALYSIS(DSC) RESULTS INDICATED THAT THE ENDOTHERMIC PEAK WAS BROADER AND SHIFTED TO LOWER TEMPERATURE IMPLYING THE PRESENCE OF DRUG. MORE THAN 90% OF DRUG OF NANOCAPSULES WAS DETECTED IN PHOSPHATE BUFFER (PH=7.4) AND37°C TEMPERATURE USING UV-VIS SPECTROPHOTOMETRY.
