Abstract: 

Introduction: It has been demonstrated that use of plasma rich in growth factors produces a dynamic surface on the implants, which accelerates the healing process. This study undertook a histologic and histomorphometric evaluation of the effect of platelet-rich growth factors (PRGF) on the amount of bone regeneration around dental implants.Materials and Methods: In this animal-intervention study, two systemically healthy Persian mixbreed dogs were selected. Three months after extraction of the dogs’ mandibular premolars on both sides, 12 implants were selected from the OIC system with a length of 10 mm and a diameter of 5 mm.Six implants were placed in the osteotomy cavities using growth factors and 6 others were placed in the osteotomy cavities without PRGF. After 12 weeks, all the implants were removed along with the surrounding bone using Trephine bur. The samples were evaluated under a light microscope subsequent to sectioning. Data were analyzed with t-test and Mann-Whitney test (a=0.05).Results: The results of the present study indicated that histologically, there was a significant increase in the amount of regenerated bone around the implants, moreover, thickness and maturity of bony trabeculae were not different in the samples containing PRGF in comparison with the control samples.Conclusion: According to the results, implant-bone interface integration rate around dental implants coated with PRGF was higher compared to the control group.

Abstract: 

Objective: Different techniques have been suggested for the repair of bone defects at the injured sites. Use of biomembranes, or application of plasma rich in growth factor (PRGF) at the site of proliferation of osteoblasts are among the suggested techniques. The current study aimed to compare the biocompatibility of human periodontal ligament fibroblasts (hPLF) cultured on Hypro-Sorb F, Pericardium and Tutodent resorbable membranes coated with PRGF.Methods: This experimental study was conducted on four resorbable membranes namely Hypro-Sorb F, Pericardium, Tutodent and Vicryl. Fibroblast cells isolated from the periodontal ligament of premolar tooth were passaged three times and 105 cells/ cm2 were cultured on membranes coated with PRGF. After 72 hours, the cells were evaluated in terms of biocompatibility and alkaline phosphatase (ALP) activity. Statistical analysis was carried out using one-way ANOVA.Results: PRGF increased cell adhesion and Tutodent membrane coated with PRGF showed the highest cell adhesion compared with Hypro-Sorb F and Pericardium membranes (p=0.005).Conclusion: PRGF increases cell viability and ALP activity of cells on biomembranes. PRGF treatment increases the adhesion of fibroblast cells to these membranes.

Abstract: 

سابقه و هدف: با توجه به اینکه رژنراسیون ضایعات استخوانی بزرگ از چالش های اصلی بازسازی فکی است، مطالعه حاضر به بررسی تاثیر استفاده همزمان از ماتریکس دمینرالیزه استخوان (Demineralized Bone Matrix) DBM، سلول های بنیادی مزانشیمال MSCs (Mesenchymal Stem Cell) و پلاسمای غنی از فاکتور رشدی (Platelet rich in growth factors) PRGF می پردازد.مواد و روشها: سلول های بنیادی با استفاده از مغز استخوان ناحیه فمور خرگوش ها استخراج و ماهیت بنیادی آنها اثبات گردید. PRGF با استفاده از 5 سی سی خون که از قلب خرگوش ها آسپیره شده بود با استفاده از دستگاه سانتریفوژ تهیه گردید. طی فرآیند جراحی، 4 نقص 8 میلی متری بوسیله فرز ترفاین در ناحیه جمجمه خرگوش ها ایجاد شد. سلول های پاساژ سوم روی DBM بارگذاری شده، 24 ساعت در انکوباتور نگهداری شدند. ترتیب جایگذاری مواد به این صورت بود: گروه DBM=1، گروه DBM+PRGF=2، گروه DBM+MSCs=3 و گروه 4=DBM+PRGF+MSCs. در هفته 6 و 12 خرگوش ها به شیوه وایتال پرفیوژن قربانی شده، آنالیز هیستومورفومتری بر روی تقاطع 5 میکرومتری با رنگ آمیزی هماتوکسیلین و ائوزین (H&E) انجام شد. تحلیل داده ها با استفاده از نرم افزار SPSS نسخه 14 و با آزمون ANOVA انجام گرفت.یافته ها: آنالیز هیستومورفومتری کلیه مقاطع در 6 و 12 هفته پس از جراحی، استخوانسازی را در گروه 1، 74.6% و 20.31% در گروه 2، 14.35% و 28.44%، در گروه 3، 17.75% و 31.33% و در گروه 4، 18.94% و 37.21% نشان داد. درصد استخوان سازی بالاتر در نقص گروه 4 مشاهده شد. این اختلاف از لحاظ آماری معنی دار نبود (P>0.05).نتیجه گیری: به نظر می رسد استفاده همزمان DBM، PRGF، MSCs موجب افزایش رژنراسیون استخوان در ضایعات 8 میلیمتری کالواریای خرگوش نسبت به استفاده از MSCs یا PRGF خواهد شد.

Abstract: 

Introduction: The aim of periodontal treatment is to regenerate periodontium. Regenerative treatments include the use of plasma that is rich in growth factors (PRGF).Materials & Methods: In a randomized clinical trial, 20 three-walled intrabony defects from five patients with moderate periodontitis were randomly assigned to three groups. Patients in the control group underwent debridement of lesions. In the first treatment group, the defects were debrided and cenomembrane was applied. The third group was treated with debridement, PRGF and cenomembrane. Measures of vertical probing depth (VPD), vertical clinical attachment level (VCAL), gingival index (GI; Sinless and Loe) and radiographic index (by digital subtraction) were made preoperatively and 6 months post-surgery. Wilcoxon signed-ranks and Chi-square tests were used for analyzing quantitative and qualitative variables, respectively.Results: All three groups showed improvements in all measures except GI. Intra-group comparison for clinical attachment level (CAL) indicated significant difference in all groups before and after surgery (P<0.05); there was no correlation in CAL among groups before surgery Intra-group comparison demonstrated significant differences in all three groups before and after surgery (P<0.001). A statistically significant difference was found in radiographic indices among the groups post-surgery (P=0.009).Conclusion: The use of PRGF was associated with improvements in all parameters but not for GI.

Abstract: 

Objective: Sperm cryopreservation results in damage to membrane integrity, sperm viability, sperm motility, and DNA structure. We aimed to evaluate the effect of plasma rich in growth factors (PRGF) on sperm parameters during the freeze-thaw process. Materials and Methods: In the first phase of this prospective study, after sperm preparation, 10 normozoospermic specimens were cryopreserved by rapid freezing with different concentrations of PRGF including 0, 1, 5, and 10% to find the optimum dose. Sperm motility and viability were assessed in this phase. In the second phase of the study, based on the results of first phase, 25 normal sperm samples were frozen with 1% PRGF. All sperm parameters including motility, viability, acrosome reaction, and DNA integrity were assessed before freezing and after thawing. Results: The rates of progressive and total sperm motility and viability were significantly higher in 1% PRGF compared to control, 5%, and 10% PRGF in the first phase (P<0. 05). Supplementation of freezing medium with 1% PRGF could significantly improve all sperm parameters including sperm motility, viability, normal morphology, acrosome integrity, chromatin structure, chromatin integrity, DNA denaturation, and DNA fragmentation in comparison with the control group. Conclusion: It appears that the supplementation of freezing medium with 1% PRGF could protect human sperm parameters during cryopreservation.

Abstract: 

Objective: This randomized clinical trial was aimed at comparing the outcomes of palatal connective tissue + Bio-Oss + PRGF versuscollagen membrane + Bio-Oss+ PRGFin the treatment of intrabny defects.Methods: Fifteen patients affected by chronic periodontitis were enrolled. Each patient had at least two intrabonydefect (³3 mm). They were randomly assigned into two groups: patients treated i) with collagen membrane + PRGF + Bio-oss (control group) and ii) with palatal connective tissue as membrane + PRGF + Bio-Oss (experimental group). Clinical and intrasurgical examinations included probing depth (PD), clinical attachment level (CAL), gingival recession (GR), defect fill (DF), alveolar crest level (AC) and defect resolution (DR) were measured at baseline and after 6 months with re-entry surgery. Statistical analysis was performed usingTwo-way Repeated Measure ANOVA and Wilcoxon.Results: After 6 months, all of the evaluated clinical parameters showed statistically significant changes from baseline within each group (p<0.05). The test group showed a significantly smaller amount of gingival recession as compared with control group (0.8 mm versus 1.7 mm respectively; P<0.05). But there were not any statistically differences in other clinical parameters between the test and control groups (P>0.05): pocket depth reduction (3.7 mm versus 3.5 mm), clinical attachment gain (1.8 mm versus 1.6 mm), alveolar crest loss (1.8 mm versus 1.7 mm), defect fill (2.3 mm versus 2.2 mm) and defect resolution (4.1 mm versus 3.9 mm).Conclusion: According to the results obtained from this study it can be suggested that clinical effects of application of palatal connective tissue as a membrane and collagen membrane, in combination with Bio-Oss and PRGF in treatment of vertical bone defects, did not have any statistical significant differences except for gingival recession that was statistically smaller in connective tissue group.
