Abstract: 

Background: Herceptin is a monoclonal antibody that is used in treating breast cancer. In this study, Herceptin is labeled with lutetium-177 and after performing all the quality control tests the therapeutic efficiency was evaluated in the mice bearing breast tumor.Methods: Herceptin was labeled with 177Lu via DOTA. The quality control tests included labeling efficiency, stability in buffer and blood serum, immunoreactivity and stability in mice were performed and therapeutic efficiency was evaluated in mice bearing breast tumor.Results: Labeling efficiency more than 90%, stability in buffer and blood serum up to 7 days 85% and 75% respectively, immunoreactivity 86% suggested that 177Lu-Herceptin could be used as a radio immunotherapy agent. The results of therapy in mice showed that the injection of complex (300 m Ci) could limit the tumor growth up to 15 days.Conclusion: By the good results of animal study, 177Lu-Herceptin could be considered as a new radiopharmaceutical agent for use in radio immunotherapy of breast cancer in humans that needs more investigation.

Abstract: 

Background: IL-6 cytokine acts as a pro inflammatory factor in the tumor microenvironment in the breast cancer. As there is a positive feedback loop between IL-6 and microRNA let-7a, which is postulated to function as a tumor suppressor in most cancers, we investigated the effects of aerobic training on microRNA let-7a expression and levels of tumor tissue IL-6 in mice with breast cancer.Methods: Twenty BALB/c mice (4-5 weeks old) were injected estrogen-dependent receptor breast cancer cells MC4-L2 and divided into two groups: tumor-training (TT) and tumor-control (TC) group. Then TT group performed progressive aerobic training for 6 weeks, 5 days per week (14-18 m/min). After tumor emersion, tumor width and length were measured by digital caliper every week.48 hours after the last exercise subjects were killed. Tissue sampling were collected and stored in -70o. Tumor tissue was homogenized and let-7a expression and IL-6 levels were accounted with Real time-PCR and ELISA Kit respectively. Statistical analysis of let-7a was conducted by the REST software. Repeated measures and independent t tests were used to assess tumor size and IL-6, respectively.Results: Tumor size and IL-6 levels were significantly decreased in TT group compared with TC group (p<0.05). MicroRNA let-7a was increased significantly in TT group following aerobic training compared with TC group (p<0.05).Conclusion: Reduction in tumor size in BALB/c mice and, followed by aerobic exercise can be attributed to the loss of inflammatory factors such as IL-6 and also up regulation of microRNA let-7a. These data provide strong support for a beneficial effect of exercise training on tumor progression in mouse model of breast cancer that may be partly mediated by its anti-inflammatory potential.

Abstract: 

Background: Breast cancer is the most common cancer in women. P53 is the most important tumor suppressor genes and genetic polymorphism of p53 is known as a risk factor for breast cancer. P53 mutation has been seen in 20-40% of breast cancer patients. DNA damage causes p53 phosphorylation and p53 accumulation in nucleus. Therefore the aim of this study is assess the evaluation of p53 Arg/Pro polymorphism at codon 72 and expression of p53 protein in breast cancer women in Yazd city.Methods: In case-control study, 104 patients with breast cancer and 104 matched controls in Yazd city were chosen. After DNA extraction, P53 codon 72 polymorphism was determined by ARMS-PCR technique and P53 overexpression was evaluated by immunohistochemistry method.Results: In breast cancer patients group, the distribution of codon 72 polymorphism for Arg/Arg, Pro/Pro and Arg/Pro is 49.04, 21.15 and 29.81 percent respectively and in control group is 21.15, 26.92 and 51.93 percent respectively. The significant difference was seen between distribution of Arg/Arg and Arg/Pro genotype in breast cancer patients and control group (P˂0.05). Also P53 negative was observed in 71.15% and p53 positive in 28.8% of breast cancer patients. Therefore, significant difference was seen between p53 positive and negative (P<0.01).Conclusion: The results of this study showed that Arg/Arg genotype is a talented genetic factor for breast cancer, while Arg/Pro genotype has protective role against breast cancer.Also the tumor tissues of breast cancer patients have shown different levels of p53 protein.

Abstract: 

Background: Genetic mutations in premalignant breast lesions may have a role in malignancy progression or influence the behavior of subsequent disease. A point mutation in estrogen receptor-a (ESR1) as A908G (Lys303®Arg) was originally involved to hypersensitive to estrogen breast hyperplasia. We detected this mutation among Iranian women with invasive breast cancer.Methods: A population-based case-control study was conducted in 150 newly diagnosed invasive breast cancer and 147 healthy control individuals to screen for presence of the ESR1 A908G mutation by using single-strand conformation polymorphism (SSCP) analysis and 33Pcycle DNA sequencing.Results: We detected the 10.7% ESR1 A908G mutation in the form of heterozygote genotype only among cancer patients (P=0.00). The allelic frequency of mutant allele AGG in codon 303 was significantly (P=0.001) higher in patients with the family history of breast cancer (28.9%) than those without the family history of breast cancer (1.9%).Conclusion: Our data suggest that ESR1 codon 303 mutations are correlated with various aspects of breast cancer in Iran. ER-α genotype might represent a surrogate marker for predicting breast cancer developing later in life.

Abstract: 

Background: Mammography is an approved screening technique which may reduce breast cancer mortality. Evidences suggest that women with high breast density are four to five times more likely to get breast cancer than women with low breast density. In order to examine the effect of several factors on mammographic density, we designed and conducted this study in our breast clinic.Methods: In this cross sectional study a structured questionnaire consisting of several variables such as age, reproductive index, BMI, OCP consumption, age of menarche, breast cup size was fulfilled. Completed data were analyzed and their correlation with breast density was studied. We used chi square test and logistic regression for determining the relationship between the covariates and density of breast tissue variable.Results: In total 428 questionnaires were reviewed. About 65% of populations (277 women) were Under the age of 50 year and 44% of them were more than 50. The results indicate that there is a significant correlation between the low age, low BMI, premenopausal status and small breast cup size with breast density in mammography (P>0.0001). But the other variables did not show significant correlation.Conclusion: High density of breast tissue in premenopausal young women with lower BMI may decrease the sensitivity of mammography. This finding insists on the importance and necessity of short interval examination and applying other diagnostic procedures in these women.

Abstract: 

Background: In many cultures, breast is a symbols of femininity, motherhood, beauty and sexual attractiveness that loss and deformity it's may lead to negative effect on sexual life.Objective: The objective of present study is to understand sexual function after mastectomy surgery in breast cancer survivors.Methods: This study has a qualitative design. Semi-structured interview have been conducted with 11 Iranian women surviving breast cancer to explore their view on sexual function after mastectomy.Results: average age of participants and husbands was 44.5±4.9 and 47.8±5.2 respectively. Average time between surgery and interview was 2.8 (rang: 1-7 years). Analysis using the Colaizzi method reveals four major themes: (1) dyspareunia, (2) hesitation in resuming sex after mastectomy surgery, (3) decrease of frequency and quality of sex, and (4) sexual considerateness.Conclusion: This study shows changes of sexual function after mastectomy surgery. In addition, understanding and cognition of sexual life among breast cancer survivors help to improve survivors' sexual wellbeing. The finding this study helps to mental healthcare professionals to design appropriate interventions in order to improve survivors' sexual life.
